蛋白质组学技术对冠心病血瘀证相关蛋白的研究

摘    要:目的用蛋白质组技术观察冠心病血瘀证病人与正常人血浆中的蛋白质变化,以期发现诊断冠心病血瘀证的标志物和揭示冠心病血瘀证的发病机制.方法采集正常健康者、冠心病心血瘀阻证病人的血浆,用双向电泳、图像分析、质谱鉴定等蛋白质组学技术测定冠心病血瘀证与正常人血浆中的蛋白变化.结果蛋白质组学研究发现冠心病血瘀证病人血浆与正常相比有3个蛋白质下调和6个蛋白质上调,胶内酶切提取蛋白进行质谱鉴定,其中冠心病血瘀证病人血浆与正常相比升高的蛋白质有免疫球蛋白、纤维蛋白原、粒酶,冠心病血瘀证病人血浆与正常相比降低的蛋白质有CD44SP.结论纤维蛋白原、粒酶有望作为诊断冠心病血瘀证的标志物.

Abstract:Objective To explore protein changes in stagnation of blood type of coronary heart disease and in normal human plasma by proteomic technique, in order to find the diagnostic biomarkers of stagnation of blood type of coronary heart disease and clarify the pathogenesis. Methods Gather the plasma of normal and stagnation of blood type of coronary heart disease. The albumin in plasma was removed. Proteomic protocol involved of 2-DE, image analysis and spectrometry detection was used to detect the change of plasma protein.Results 3 protein decreased and 6 protein increased in blood plasma of stagnation of blood type of coronary heart disease, compared with normal plasma, the fommer included fibrinogen,granzyme,immunoglobulin and the latter was CD44SP.Conclusion Fibrinogen, granzyme might be developed to be diagnostic biomarkers to stagnation of blood type of coronary heart disease.
