实验性糖尿病大鼠视网膜微血管变化的动态研究

摘    要:目的 :了解糖尿病大鼠早期视网膜毛细血管变化情况。 方法 :将 3 0只Wister大鼠分为实验组与正常对照组。实验组用链脲佐菌素 (Streptozocin ,STZ)诱发糖尿病。并于糖尿病发病后 4、6和 12周观察两组视网膜毛细血管的变化情况 ,同时监测两组大鼠的体重、血糖和糖化血红蛋白 (HbA1c)。 结果 :糖尿病大鼠血糖持续高于 19mmol/L以上 (P <0 0 0 1) ,HbA1c明显高于对照组 (P <0 0 5 ) ,体重明显低于对照组 (P <0 0 5 )。视网膜血管铺片见 :糖尿病 4周 ,视网膜毛细血管无明显异常改变 ;糖尿病 6周 ,开始出现血管壁细胞数目减少 ;糖尿病 12周 ,血管壁细胞数进一步减少 ,并出现毛细血管形态的异常。对照组在各实验时点 ,视网膜毛细血管无异常改变。 结论 :糖尿病大鼠视网膜微血管最早期的改变是毛细血管壁细胞数目的减少 ,然后才发生血管形态学的异常。

Abstract:Obective:To observe the early stage change of retinal micrangium in diabetic rat. Methods: Thirty Wister rat were used in this study for two groups, experiment and control. The rats in experiment group were induced diabetes with streptozocin. The weight, fasting blood glucose (FBG), HbA 1c , and the morphology of retinal micrangium were observed in the 4th ,6th and 12th weeks after the occurrence of diabetes. Results: FBG of diabetic rats was above 19mmol/L in all course of diabetes ( P <0.001). HbA 1c of diabetic rats was significantly higher than that of the control ( P <0.05), but the weight of diabetic rats was markedly lower than that of the control ( P <0.05). The morphological examination shown that the abnormal of retinal micrangium was not found in 4th weeks after diabetes. The decreased of cell number in retinal micrangium was seen in 6th weeks after diabetes. In 12th weeks after diabetes, besides the number of cells in retinal micrangium reduced, the abnormal of morphology was also observed. During the experimental period, the abnormal in retinal micrangium was not seen in control group.Conclusions: The decreased of cell number in retinal micrangium is the early stage change in diabetic rats, then the changes of morphology take place.
