当归注射液对免疫诱导再生障碍性贫血小鼠骨髓细胞周期蛋白D2表达的影响

摘    要:目的:探讨当归注射液对免疫诱导再生障碍性贫血(AA)小鼠骨髓有核细胞增殖周期及其细胞周期蛋白D2表达的影响。方法:雌性Balb/c小鼠,随机分为3组:①正常对照组:未经特殊处理。②模型对照组:经60Co6Gyγ-射线亚致死量全身照射,4 h内由尾静脉输入取自DBA/2小鼠胸腺、淋巴结混合细胞,制成AA模型,于制模当天即给予腹腔注射无菌生理盐水,每天10 ml.kg-1,连续12 d。③当归注射液组:每天予当归注射液10 ml.kg-1,腹腔注射,连续12 d。于第12 d每只小鼠称重后,采集尾静脉血测外周血白细胞计数,后断颈处死小鼠,取出双侧尺骨及股骨,冲出骨髓单个核细胞,计数。并行组织学切片,粒单系祖细胞培养及骨髓有核细胞G0/G1期细胞百分率及细胞周期蛋白D2的检测。结果:与正常对照组比较,模型对照组造血细胞明显减少,粒单系祖细胞计数明显减低,骨髓有核细胞G0/G1期细胞百分率明显增加,而细胞周期蛋白D2表达水平明显降低,当归注射液组能增加骨髓单个核细胞计数及粒单系祖细胞计数,明显降低G0/G1期细胞百分率,提高细胞周期蛋白D2的表达水平。结论:当归注射液可能通过刺激细胞周期蛋白D2的表达促进造血细胞的增殖。

Abstract:Objective: To investigate the influence of angelica sinenisis injection on the percentage of cells in G_(0)/G_(1) and expression level of cyclin D_(2) of bone marrow mononucleate cells(MNC) in immune-induced aplastic anemia(AA) mice.Methods: The female Balb/c mice were divided into 3 groups.Group A was normal control.Group B had been generally irradiated by sub-lethal dosage ~(60)Co 6Gyγ-ray,then injected through tail vein with mixed lymphocytes of thymus gland and lymphoid node of DBA mice in 4 hours.After establishing AA models,each mouse was intraperitoneally injected with sterile saline 10 ml·kg~(-1)·d~(-1) for 12 days.Mouse from group C was intraperitoneally injected with angelica sinenisis injection 10 ml·kg~(-1)·d ~(-1)for 12 days for an intervention treatment.All mice were killed by cutting neck on the 12th day,ulnas and femur were taken out.The bone marrow histology section were observed.The number of femur MNC was measured.Meanwhile,The technique of culture of CFU-GM was used and the percentage of cells in G_(0)/G_(1) and cyclin D_(2) expression level in bone marrow was measured by the flow cytometor analysis system.Results: Compared with group A,the number of MNC,the expression levels of cyclin D_(2) obviously was reduced(P<0.01),but the percentage of G_(0)/G_(1)cells in bone marrow significantly increased(P<0.01)in group B.In group C,the number of MNC,the level of cyclin D_(2) was increased(P<0.05) and the percentage of G_(0)/G_(1)cells in bone marrow was reduced(P<(0.01).) Angelica sinenisis injection could obviously promote the proliferation of CFU-GM.(Conclusion:) The results suggest angelica sinenisis injection could promote the proliferation of bone marrow in immune-induced AA mice by increasing the expression level of cyclin D_(2).
