HSP70在发绀型先天性心脏病人心肌组织中的表达

摘    要:目的: 探讨发绀型先天性心脏病人心肌细胞中HSP70的表达及其心肌保护作用。方法:非发绀型先天性心脏病人12例(A组),发绀型先天性心脏病人12例(B组)。两组病人于阻断主动脉前、阻断主动脉后20 min及开放主动脉后20 min,取右房心肌标本,分别进行免疫组化法检测HSP70的含量及化学比色法检测SOD活性、MDA的含量。结果:心肌细胞中HSP70的含量在手术过程中无明显变化(P>0.05);B组HSP70的含量明显低于A组(P<0.05)。手术过程中B组SOD活性明显降低,MDA含量明显高于A组(P<0.05)。结论:发绀型先天性心脏病人心肌细胞中HSP70呈低水平表达与其抗缺血/再灌注损伤作用减弱有关。

Abstract:Objective To evaluate the expression and myocardial protection of heat shock protein-70(HSP70) in the myocardial cells of cyanosis congenital heart diseases. Methods The study enrolled 24 patients with congenital heart disease: 12 cases of non-cyanosis (group A), and the other 12 cases of cyanosis (group B). During the operation the samples of myocardium in the right atrium were attained at 0 min ischemia, 20 min ischemia,and 20 min reperfusion. They were used to evaluate the content of HSP70 by immunohistochemistry and activation of SOD and content of MDA by chemochromatometry.Results The content of HSP70 in myocardial cells during the operation was not significantly different in both groups (P>0.05). The content of HSP70 significantly decreased in group B compared with that of group A (P<0.05). The activity of SOD significantly decreased and the content of MDA significantly increased in group B during the operation (P<0.05).Conclusion Decreased expression of HSP70 is associated with the decreased tolerance to ischemia-reperfusion injury in myocardial cells of cyanosis congenital heart disease.
