福辛普利、普伐他丁对实验性糖尿病大鼠心肌的保护作用

摘    要:目的 :探讨福辛普利及普伐他丁对糖尿病大鼠心肌的保护作用。方法 :将 30只糖尿病大鼠随机分为福辛普利治疗组 (福辛普利组 )、福辛普利加普伐他丁联合治疗组 (联合组 )及糖尿病未治疗组 (对照组 ) ,每组 10只。用福辛普利 [15mg (kg·d) ]和普伐他丁 [2 0mg (kg·d) ]进行治疗 ,另取 10只正常大鼠作为空白对照 (正常组 )。观察糖尿病大鼠用福辛普利及普伐他丁治疗前后心肌细胞间质中层粘连蛋白、纤维连接蛋白和胶原含量以及心肌细胞超微结构的改变。结果 :经 5周治疗后 ,层粘连蛋白在正常组、联合组及福辛普利组大鼠心肌组织中的表达呈阳性反应 ,而对照组呈强阳性反应 ;纤维连接蛋白在正常组大鼠心肌组织中的表达呈弱阳性反应 ,对照组呈强阳性反应 ,福辛普利组和联合组中呈阳性反应 ;正常组心肌组织中的胶原表达为弱阳性改变 ,对照组呈强阳性改变 ,福辛普利组和联合组呈阳性改变 ;各组心肌细胞超微结构病理改变以糖尿病未治疗组较重 ,联合组改变最轻 ;联合组治疗 5周后血脂明显下降 ,福辛普利组及对照组 5周后血脂无明显下降。结论 :应用福辛普利、普伐他丁可延缓实验性糖尿病大鼠心肌细胞和细胞间质的病理学进展

Abstract:Objective: To study the heart protective effect of Fosinopril and Pravastatin on diabetic rats by observing the changes of the myocardium pathological and contents in myocardial collagen(CL),laminin(LN) and fibronectin(FN) in before and after of treatment in diabetic rats. Methods: Diabetic rats were treated with Fosinopril (15 mg·kg -1 ·d -1 ) and Pravastatin (20 mg·kg -1 ·d -1 ) for five weeks, the protective effects of Fosinopril and Pravastatin on heart cell structure and contents in myocardial collagen. Laminin and fibronectin were evaluated. Results: The microscopic findings showed after Fosinopril and Pravastatin treatment for five weeks, the heart pathological changes and contents in myocardial CL, LN and FN were markedly reduced as compared with those in treatment by Fosinopril only and untreated diabetic rats and the changes in the group of untreated were rather obvious. Conclusion: Treatment with Fosinopril and Pravastatin reduces the injure of the heart and prevent the structure of heart pathological progress in diabetic rats.
