摘    要:目的:优选大孔吸附树脂纯化头风宁滴丸中菊花的工艺条件。方法:采用高效液相色谱法,以绿原酸比吸附量、洗脱率为指标,对大孔吸附树脂型号、上样量、上样浓度、洗脱剂浓度及用量进行考察。结果:最佳工艺条件为预处理后药液浓度为0.075g·mL-1(以菊花药材计),通过HPD600型大孔吸附树脂柱,上样量为1.2g(生药)/mL树脂,以水洗至流出液近无色,继以4倍树脂体积70%乙醇洗脱。结论:该优选工艺可以较好地纯化富集绿原酸,达到"去粗存精"的纯化效果。

Abstract:OBJECTIVE: To optimize macroporous adsorptive resins to purify Chrysanthemun morifalium in Toufengning dropping pills. METHODS: The type of macroporous resin, the maximum amount of physic liquor, the concentration of physic liquor and the concentration of elution solvent were optimized with the adsorption quantity and elution rate of chlorogenic acid as indexes with HPLC. RESULTS: The optimal conditions were as follows: the concentration of the pretreated physic liquor (chrysanthemum) was 0.075 g·mL^-1, the maximum amount of physic liquor (C. morifalium) was 1.2 g·mL^-1resin, which was sieved through HPD600 macroporous resin column, washed with water until appearing of colorless effluent, followed by elution with 70% ethanol (at a volume of 4-fold that of resin). CONCLUSION: The optimized technology is effective for purifying and enriching the chlorogenic acid, which achieve the purpose of purification.
