| 摘 要： | 目的 探讨不同肝癌细胞株中转录因子FOXM1（Forkhead Box M1）及核转录因子NF-κB（Nuclear factor kappa B）的表达强度及意义.方法采用Western blot、RT-PCR法,检测不同肝癌细胞株（QGY-7703、BEL-7402、SMMC-7721）中FOXM1及NF-κB在蛋白及mRNA水平的表达情况.结果 在3株肝癌细胞中FOXM1及NF-κB的表达无论是mRNA水平还是蛋白水平差异均有统计学意义,细胞株（QGY-7703、BEL-7402）中FOXM1及NF-κB的表达同时都明显高表达于细胞株SMMC-7721.结论 肝癌细胞株中FOXM1蛋白及mRNA表达与NF-κB表达趋势一致,提示FOXM的表达可能与NF-κB的活化有关.

| Abstract: | Objective To investigate the expression and significance of Forkhead Box M1 and nuclear factor kappa B in human liver carcinoma cells. Methods The expression of Forkhead Box M1 and nuclear factor kappa B protein and micro - RNA in hepatocellular carcinoma cell lines （QGY -7703, BEL- 7402 and SMMC -7721 ） were detected with the application of Western blot analysis and RT- PCR analysis, respectively. Results The expression levels of Forkhead Box M1 and nuclear factor kappa B varied significantly among three cell lines. Western blotting and RT - PCR showed that hepatocellular carcinoma （ QGY - 7703 and BEL - 7402） cells expressed higher FOXM1 and NF - KB levels than SMMC - 7721 （ P 〈 0. 01 ）. Conclusion FOXM1 protein and mRNA expression in human liver cancer cells and the NF - KB expression trend is consistent, prompt FOXM1 expression is associated with the activation of the NF - KB. |
