摘    要:应用整装细胞电镜技术结合选择性抽提方法,观察正常人外周血单核细胞的细胞骨架系统,并应用抗α管蛋白单克隆抗体和抗肌动蛋白抗体—胶体金免疫标记技术,分别对其胞浆中微管、微丝进行了定性研究。

Abstract:Cytoskeletal framewok prepared by the selective extraction method has been observed withwhole mount cell electron microscopy.Cytoskeleton prepared by Triton X-100 treatment from hu-man periphral blood monocytes appeared in the examination by means of microscopy as a highly or-ganized and interconnected three-dimensional matrix of different fibrous elements.Since such cy-toskeleton are open membrane-free system,individual fibrous organization can be identified byspecfic antibodies.An indirect immunogold procedure with assay of monoclonal anti-tubulin oranti-actin antibodies was applied to visualize tubulin or actin containing structure.By further ex-traction with 0.25M(NH_4)_2SO_4 to remove the microtubules and microfilaments,only the salt-re-sistant intermediate filaments lattice were left.
