参麦注射液对大鼠海马神经细胞凋亡的影响

摘    要:目的以缺氧/缺糖再给氧为模型,观察参麦注射液对神经细胞凋亡的抑制作用。方法流式细胞术检测凋亡细胞百分率,激光共聚焦扫描显微镜检测细胞内Ca2+浓度,荧光显微镜观察细胞形态学变化和坏死百分率。结果缺氧/缺糖5h后再给氧可诱导神经细胞凋亡和细胞坏死,并显著增加细胞内Ca2+浓度和LDH的释放。与对照组相比有显著性差异(P<0.01)参麦注射液能显著降低神经细胞凋亡及坏死的百分率,降低细胞内Ca2+浓度,减少LDH的释放,其作用随剂量增加而增加。结论参麦注射液具有拮抗海马神经细胞凋亡的作用,这种作用可能与其能降低细胞内Ca2+浓度有关。

Abstract:Objective To study the protect effects of Shen Mai injection(SHM)on apoptosis induced by hypoxia /hyp oglycemia and reoxygenation in cultured rat hippocampal neurons.Methods Apoptosis were measured by flow cyto metry;intracellular free calcium c oncen-tration([Ca 2+ ]i)was measured with confocal laser scanning microscopy;Morphological changes and neuronal necrosis were observed with flu-orescence microscope.Results Hypoxia /hypoglycemia cultures for5hour and reoxygenation induced neu ronal apoptosis and significantly increased neuronal necrosis,signi ficantly increased neuronal[Ca 2+ ]i and the leakage of LDH.The effect we re increased with the exlending time of reoxygenation.SHM might have sig nificantly decrease the percentage of neuronal apoptosis and necrosis,reduce neuronal[Ca 2+ ]i and the leakage of LDH.Conclusion SHM had effect of anti-neuronal apop tosis.This effect might be related t o decrease intracellular free calcium concentration.
