血红素氧合酶-1高表达抗兔心肌缺血再灌注损伤研究

摘    要:目的探讨血红素氧合酶-1(HO-1)高表达抗兔心肌IR损伤的效果及其作用机制。方法32只兔随机均分为单纯IR组与HO-1诱导组,分别预先给予生理盐水5mL及血晶素25mg/kg腹腔内注入。24h后,开胸结扎冠脉左前降支缺血40min,松开再灌注4h,测定心肌中HO-1表达及活性、中性粒细胞浸润、丙二醛(MDA)含量、心肌梗死范围及心肌细胞凋亡指数。结果与单纯IR组比较,HO-1预诱导组心肌中HO-1表达及活性均明显升高(P〈0.01),中性粒细胞浸润及MDA含量明显降低(P〈0.01),心肌梗死范围及心肌细胞凋亡指数均显著减少(P〈0.01)。结论HO-1可通过抗炎、抗氧化等作用减轻急性心肌IR损伤。

Abstract:[Objective] To explore the effects and mechanism of heme oxygenase-1(HO-1)overexpression protection of rabbit heart from ischemia reprfusion(IR)injury.[Methods] 32 rabbits were randomly divided into simple IR group and HO-1 induced group,each rabbit was intra-peritoneal injections of 5 mL saline in simple IR group and 25mg/kg hemin in HO-1 induced group respectively.24 hours later,rabbit heart underwent ischemia for 40 minutes by ligatin left anterior descending coronary artery and reperfusion for 4 hours.The myocardial HO-1 expression and HO-1 activity,polymorphonuclear neutrophils(PMN)infiltration,MDA content,infarct size and the apoptosis index(AI)in both groups were detected.[Results] The amount and activity of myocardial HO-1 of HO-1 induced group were significantly higher than that of simple IR group(P <0.01 respectively).While the myocardial PMN infiltration,MDA content,infarct size and the AI of HO-1 induced group were much lower than that of simple IR group(P <0.01,respectively).[Conclusion] HO-1 can reduce myocardial IR injury by means of anti-oxidative and anti-inflammatory properities.
