摘    要:目的探讨急性肺血栓栓塞大鼠的肺组织中儿茶酚胺代谢相关酶类氧甲基转移酶(COMT)和单胺氧化酶(MAO)的表达变化。方法建立大鼠急性肺血栓栓塞模型,分别在血栓栓塞后1、8、24和48 h采取心脏穿刺法取血,用半定量RT-PCR法和Western blot法检测COMT、MAO-A及MAO-B的mRNA和蛋白表达;用HPLC法检测血清中儿茶酚胺水平。结果在急性肺栓塞的不同时间点,COMT、MAO-A及MAO-B的mRNA和蛋白水平均逐渐降低,而血清中肾上腺素(EPH)和去甲肾上腺素(NE)呈逐渐升高趋势。结论急性肺栓塞大鼠的血清儿茶酚胺水平升高源自肺组织内的降解减少。本研究部分阐明了急性肺栓塞导致肺动脉高压的分子机制。

Abstract:Objective To study the expression of catecholamine metabolism related enzymes COMT and MAO in the lung tissues of a rat acute pulmonary embolism(PE) model.Methods We constructed an acute PE model.The blood sample was collected at 1 h,8 h,24 h and 48 h.mRNA level changes of COMT,MAO-A and MAO-B were identified by semi-quantitative RT-PCR,and the protein level changes of COMT,MAO-A and MAO-B were validated by western blot.The serum concentrations of catecholamine were determined by HPLC methods.Results At different time points,the mRNA and expression of COMT,MAO-A and MAO-B gradually decreased after acute PE.However,the serum concentrations of epinephrine(EPH) and norepinephrine(NE) increased gradually as compared with that in controls.Conclusion The serum concentrations of catecholamine increased after acute PE because its degradation was suppressed.The study of their expression changes may be helpful to find the molecular mechanisms of pulmonary artery hypertension involved in acute PE.
