汉坦病毒核衣壳蛋白抗原多肽片段的合成与应用

目的 以合成肽为抗原建立新型特异性检测抗汉埋病毒IgG和IgM的ELISA。方法 用多肽合成仪合成汉坦病毒核衣原壳蛋白（aa17-66)50个氨基酸残基的核心序列，以此为抗原，通过实验发现这一序列具有较强的抗原性，首次建立了以合成肽为抗原的新型特异性检测抗汉坦病毒IgG和IgM的ELISA，测定肾综合征出血热（HFRS）患者血清23份，乙型肝炎患者血清9份，正常人血清11份。结果 乙型肝炎患者血清和正常人血清均为阴性，20份HFRS血清IgG阳性，21份HFRS血清IgM阳性，本方法的相对敏感性为86％和91％，相对特异性为100％，符合率为93％和95％。结论 本法特异性强，灵敏度高，简便易行，是适于基层医疗单位的诊断方法。

Preparation of major antigenic epitope of Hantaan virus nucleocapsid protein and its applications

AIM To establish a new specific serological diag nostic method to test HFRS. METHODS Epitope (aa17 66) of nucleocapsid protein was synthesized by solid phase peptide synthesis. Then the synthetic peptide was applied as an antigen to test the specific antibody (IgG, IgM) by ELISA. Twenty three sera from HFRS patients,9 from HBV patients and 11 from healthy people were detected. RESULTS All sera from HBV patients and healthy people were negative, 20 from HFRS patients for anti HV IgG positive and 21 sera from HFRS patients for anti HV IgM positive. Sensitivity of the results was 86% and 91% respectively; the specificity was 100%, and the coincidence rate was 93% and 95% respectively. CONCLUSION ELISA has been established to detect specially the anti HV IgM and IgG. It is a simple, fast, highly specific and sensitive method suitable for early diagnosis of HFRS infected people.
