超声波终止小鼠早孕的组织学研究

摘    要:本文报道了一种应用治疗剂量超声抗鼠早孕的方法。结果显示:用12W/cm~2、16W/cm~2、20W/cm~2三组声强辐照小鼠妊娠第7天在体胚胎着床点的组织学急性改变为胎盘出血,电镜下见胎盘和胚胎细胞多数受到严重损伤和细胞质膜失去连续性、细胞核碎裂等;子宫内膜和平滑肌细胞出现线粒体肿胀、内膜细胞的胞质内有的局灶性坏死.受辐照小鼠继续妊娠至第16天时处死发现受辐照的胚胎100%死亡,光镜下见胎盘、胚胎结构解体、塌陷、吸收,细胞核多数溶解,部分呈核淡染、固缩,碎裂或仅存空的子宫腔,子宫内膜增生,平滑肌无明显改变,电镜下见胚胎和胎盘失去正常细胞结构,多为细胞碎片,子宫内膜细胞和平滑肌细胞未见明显异常。结果表明,孕体较母体组织对超声更敏感,更易受损。同时,也说明超声波抗鼠早孕是可行的。

Abstract:This paper reports a new method of contragestion of appling the ultrasound irradiation of therapeutic intensity.The results revealed that the acute histological change of implantation sites of 7th day mouse embryos after ultrasound exposure with an intensity of 12W/cm2, 16W/cm2, or 20W/cm2 was placental hemorrhage, under electron microscope most placental and embryonic cells were revealed to be seriously damaged, such as lose of continuity of cytoplasmic membrane and fragmentation of cell nucleus. Mitochondrial swelling of endometrial and myometrial cells and local necrosis in cytoplasm were also observed. At sixteenth day of gestation, the animals were sacrificed. It was found that 100% of the embryos were killed. Under optical microscope, disintegration, collapse and absorbtion of placental and embryonic structrues were observed and it was laso found that most cell nuclei dissolved, some nuclei showed shallow-stained, concentration and fragmentation. In some animals, only empty uterine cavites were found. Uterme endometrium proliferated with no obvious change in the smooth-muscles. Under electron microscope, the cells of the embryos and the placenta was found to have lost normal structures, while uterine endometrium and myometrium displayed no obvious abnormality. This result proves that the conceptus are more sensitive and injured to ultrasonic irradiation of thetherapeutical dosage than dam's uterus, that it is feasible for early pregnancy termination of mouse with ultrasound.
