摘    要:目的 观察CpGODN对日本血吸虫感染小鼠后不同时期Th1/Th2细胞因子表达水平的影响 ,探讨CpGODN介导Th1/Th2免疫偏移对控制血吸虫虫卵肉芽肿的作用。方法 取小鼠感染后 4、8、12和 16周的脾淋巴细胞经CpGODN和N CpGODN处理后 ,培养 72h ,用ELISA双抗体夹心法测定培养上清中IFNγ和IL - 4表达水平的动态变化。结果 CpGODN能显著上调IFNγ的表达水平 ,下调IL - 4的表达水平 ,而N -CpGODN对IFNγ及IL - 4表达水平无显著影响。CpGODN处理后小鼠脾淋巴细胞Th2分化指数明显减小。结论 CpGODN可以调节Th1/Th2细胞因子的表达水平 ,显著上调Th1型细胞因子水平和下调Th2型细胞因子水平 ,介导小鼠Th1型细胞免疫优势应答 ,为控制日本血吸虫慢性肉芽肿病变提供了一种新途径。

Abstract:To observe the effect of CpG ODN on levels of Th1/Th2 cytokine expression levels in the mice infected with S.japonicum and the role of CpG ODN to mediate Th1/Th2 immune deviation and control granuloma formation.Spleen lymphocyte cultures were prepared from the mice infected with S.japonicum at 4, 8,12 and 16 week after infection.The cultures were incubated with CpG ODN and N CpG ODN for 72 hours.And ELISA kit was used for the quantitative determination of IFNγ and IL 4 in the spleen lymphocyte culturesm.It was found that CpG ODN could significantly up regulate the expression of IFNγ and down-regulate the expression of IL 4.But N CpG ODN could not affect the expression of IFNγ and IL 4 (P>0.05).The Th2 differentiation index of the spleen lymphocyte was significantly reduced with the effect of CpG ODN.It concludes that CpG ODN could regulate Th1/Th2 cytokine expression levels,up regulating the levels of Th1 type cytokine and down regulating the levels of Th2 type cytokine.These findings suggeste that CpG ODN as a mediator of Th1optimal immune response could be used as a novel potential approach to control the egg granuloma formation of S.japonicum.
