蕈样肉芽肿患者皮损中朗格汉斯细胞的迁移研究

摘    要:目的:探讨蕈样肉芽肿(MF)患者皮损中皮肤淋巴细胞相关抗原(CLA)、CD62L、共刺激分子(CD40、CD80、CD86)分布及其在树突状细胞(DC)上的表达。方法:将MF皮损作冰冻切片,行单克隆抗体免疫组化染色。结果:肿瘤组织真皮及表皮内可见大量CLA、CD62L、主要组织相容性复合体(MHC)Ⅱ、共刺激分子表达,CLA的表达细胞明显多于CD62L,由斑块期到肿瘤期,表皮内CLA表达数目明显减少,真皮中CLA、CD62L表达增强。CD83^+DC能表达强的共刺激分子,在基底层、真皮和血管内可见CD83^+DC表达CD62L,但未成熟DC常表达CLA。结论:MF皮损中朗格汉斯细胞发生迁移,参与了抗肿瘤免疫反应。

Abstract:Objective: To identify the expressive situation of cutaneous lymphocyte-associated antigen(CLA), CD62L,MHC II and co-stimulate molecule in the skin infiltrates of MF patients and dyeing expression of those molecules on the surface of dendritic cells(DCs). Methods: Mab and immunohistochemistry were used to detect the expression of special antigen on the surface of DC in situ. Results: The equal numbers of CLA, CD62L, MHC II and co-stimulate molecules were expressed in the skin infiltrate of MF. The numbers of CLA+ cells were more than CD62L+ cells, but from plaque stage to tumor stage, the numbers of CLA+ cells in the epidermis decreased and the expression of CLA and CD62L were enhanced in the dermis. CD83+ DCs could strongly expressed co-stimulate molecules. The CD83+ DCs expressing CD62L were easily seen in the basal layer, in the dermis and in the vascular, but immature DCs expressed often CLA molecules. Conclusion: Langerhans cells in the skin lesions could migrate and induce an anti-tumor immune response in MF.
