BMSCs/PRF复合载蛇床子素支架材料修复兔下颌骨缺损的实验研究

摘    要:目的:探讨BMSCs/PRF复合载蛇床子素支架材料修复兔下颌骨缺损的实验研究的可行性。方法:36只新西兰大白兔随机分为3组,每组12只,双侧下颌骨制备缺损模型。A组植入BMSCs/PRF复合载蛇床子素支架材料;B组植入BMSCs复合载蛇床子素支架材料,C组单植入纳米羟基磷灰石支架材料。分别于术后2、4、8、12周处死实验动物,制备组织标本,行大体观察、影像学分析、HE染色、扫描电镜检测,并对影像学分析值及成骨情况加以评价,对分析数据结果使用SPSS17.0统计软件进行分析,P〈0.05有统计意义。结果:影像学检查及组织学染色显示A组骨缺损处愈合程度,成骨速度及其质量明显优于B组C组;扫描电镜显示A组该复合材料与组织相容性好,无炎症刺激反应;统计牙CT分析数据及新生骨面积,A组的成骨情况明显优于B组与C组(P〈0.05)。结论:BMSCs/PRF复合载蛇床子素支架材料有明显骨诱导作用,是一种新型复合材料,可望成为临床应用中修复颌骨缺损的新型材料。

Abstract:Objective: To investigate the feasibility of the BMSCs / PRF compound with the Osthol scaffolds for repair the mandibular defects of the rabbit. Methods: 36 New Zealand white rabbits were randomly divided into 3 groups. Bilateral mandibular defect model was prepared. A: group of implanted MSCs / PRF composite scaffolds Osthol; B: group implanted BMSCs composite scaffolds containing Osthol, C: group of single nano--hydroxyapa- tite scaffolds implanted. All experimental animals were sacrificed after 2,4,8,12 weeks to prepare tissue samples. The observation in general and radiographic analysis were carried out. HE dyeing, SEM detecting, and analysis of the value of the Imaging and the bone were performed. Results: In vivo imaging and histological staining showed that bone defect bone quality of a group was significantly better than those of other groups. SEM showed the well composite material and tissue compatibility, no inflammation'reaction By measuring and analyzed the dental CT da- ta and new bone area, the A group bone was obviously better than the other groups (P〈0.05). Conclusion: The BMSCs / PRF compound with the Osthol scaffolds for repair the mandibular defects of the rabbit has obvious induc- tion of bone. It is a new composite material, expected to become the clinical application of new materials to repair the mandibular defects.
