牛磺酸对胚胎中脑细胞移植后的保护作用

摘    要:①目的 观察牛磺酸对帕金森病 (PD)模型大鼠胚胎中脑细胞移植术后移植细胞的存活及生长的影响。②方法 应用 6 羟基多巴胺 (6 OHDA)制备偏侧损毁的PD模型大鼠 ,将胚胎腹侧中脑 (VM )细胞悬液或生理盐水 (NS) (对照组 )立体定位移植到模型大鼠的损毁侧纹状体中 ,VM移植大鼠于术后侧脑室分别注射NS或牛磺酸 ,5 6d后通过免疫组织化学酪氨酸羟化酶 (TH)染色观察纹状体中多巴胺 (DA)能神经元 (TH阳性细胞 )的存活及生长情况。③结果 行胚胎中脑细胞移植术后 ,在PD模型大鼠的损毁侧纹状体中可检测到移植成活的DA能神经元 ,侧脑室注射牛磺酸组比侧脑室注射NS组细胞成活数目明显增多 (F =5 0 8.5 ,q =3.9,P <0 .0 1)。而在对照组模型大鼠的损毁侧纹状体中未检测到DA能神经元。④结论 牛磺酸可有效改善移植细胞的存活条件 ,并促进移植细胞的生长。

Abstract:Objective\ To study the effect of taurine on the living and growth of transplanted cells in Parkinson disease (PD) rat models after fetal mesencephalon cells transplantation. \ Methods\ The rat models of hemiparkinsonism were prepared with 6 hydroxydopamine (6 OHDA). Cell suspension of ventral mesencephalic (VM) tissue of fetal rats or normal saline (NS) (control group) was stereotaxicly transplanted into striatum of lesioned side of the rat models. NS or taurine was injected into the intracerebral ventricle of the PD rat models transplanted with VM. Fifty six days later, using immunohistochemical staining technique of tyrosine hydroxylase (TH), we observed the living and growth of dopaminergic (DA) neurons (TH positive cells) in striatum of the rat models. \ Results\ Survival transplanted dopaminergic neurons could be observed in striatum of lesioned side of PD rat models after fetal mesencephalon cells transplantation. The number of dopaminergic neurons after treatment with taurine was extremely higher compared with those treatment with NS ( F=508.5,q=3.9,P <0.01). While dopaminergic neurons could not be observed in striatum of lesioned side of the control rat models. \ Conclusion\ Taurine could efficiently improve the living condition of transplanted cells and promote them to grow.
