高效液相色谱法测定人血清中盐酸去甲万古霉素含量

目的建立反相高效液相色谱方法测定人血清中盐酸去甲万古霉素含量,并用于临床患者血药浓度监测,为个体化给药提供依据,减少不良反应发生。方法采用Synergi4μ Fusion-RP80A（250×4.6mm,4μm）色谱柱;磷酸二氢钾缓冲盐（0.02%三乙胺,用磷酸调pH=3.10）-乙腈（93：7,V/V）为流动相;流速为1.0mL.min-1;紫外检测波长：210nm;柱温：30℃;进样量：30μL。结果盐酸去甲万古霉素血清浓度与峰面积在2.0～20.0μg.mL-1范围内线性关系良好,其回归方程为：Y=30187X-3485.3,r=0.9993;低、中、高浓度的平均相对回收率分别为96.61%,95.79%,101.09%;日内精密度分别为6.21%,6.35%,10.74%,日间精密度分别为10.19%,5.89%,6.09%。结论该方法简便、快速,灵敏度高,重复性好,回收率稳定,可用于临床盐酸去甲万古霉素人体血药浓度监测及相关研究应用。

Determination of norvancomycin hydrochloride in human serum by HPLC

OBJECTIVE To develop a method for the determination of norvancomycin hydrochloride concentration in human serum by HPLC.METHODS The Synergi 4μ Fusion-RP 80A （250 mm×4.6 mm,4μm） was used,the mobile phase was potassium dihydrogen phosphate buffer （0.02%TEA,pH=3.10）-acetonitrile （93∶7,v/v） and flow rate was 1.0 mL·min-1;the detecting wavelength of UV was at 210 nm;the column temperature was 30℃;the sampling volume was 30μL.Results：Standard curve was liner in the range of 2.0～20.0 μg·mL-1;The linear equation of norvancomycin hydrochloride was Y=30187X-3485.3,r=0.9993;As for norvancomycin hydrochloride,the average recovery rate of three concentrations were 96.61%,95.79%,101.09%;The intra-day RSD was6.21%,6.35%,10.74% and the inter-day RSD was 10.19%,5.89%,6.09%.CONCLUSION The operation was simple and convenient.It is applicable to determinate norvancomycin hydrochloride concentration in human serum for clinical practice and other fields.
