连翘干燥叶片高质量DNA的提取方法研究

目的：探讨不同DNA提取方法对连翘干燥叶片的DNA质量及RAPD—PCR标记结果的影响，找出提取连翘总DNA的最佳方法。方法：分别用常规CTAB法、高盐低pH法和改良CTAB法提取连翘总DNA．分别进行琼脂糖凝胶电泳、紫外吸收A260／A280和RAPD—PCR分析，通过比较找出提取连翘总DNA的最佳方法。结果：以CTAB法为基础的改良CTAB法可获得较高质量的DNA进行PCR扩增。结论：改良CTAB法是一种分离高质量完整DNA的简便、快速方法。

Isolation of High Quality Genomic DNA From Dried Leaves of Forsythia Suspense (Thunb.) Vahl

Objective：To explore the quality of genomic DNA and its influence on RAPD-PCR of Forsythia suspense （Thunb.） Vahl with three extraction methods of DNA. Find out the most suitable extraction method of DNA from Forsythia suspense （Thunb.） Vahl. Methods：The cetytrimethyle ammonium bromide （CTAB） method,low PH extraction medium with high salt and modified CTAB method were used to extract genomic DNA from dried leaves of Forsythia suspense （Thunb.） Vahl. The DNA samples obtained were tested by 0.8% agarose gel electrophoresis,ultraviolet absorption at the ratio of A260/A280 and RAPD-PCR respectively. The most suitable extraction method of DNA from Forsythia suspense （Thunb.） Vabl was finally found out. Results：Modified CTAB method acquired the DNA from Forsythia suspensa（Thunb.） Vahl of the higher quantity to proceed PCR. Conclusion ： Modified CTAB is a simple and fast method for separating complete DNA in high quantity.
