5-羟色胺转运体基因与中国男性吸烟行为的关联研究

目的:探讨5-羟色胺转运体基因启动子区多态性(5-HTTLPR)与中国成年男性吸烟行为之间的关系。方法:采用吸烟行为相关因素问卷、艾森克人格问卷(EPQ)及尼古丁依赖评定问卷(FTND)评定中国成年男性不吸烟者及吸烟者共1012例,收集血标本采用聚合酶链反应技术检测5-HTTLPR的多态性,使用SPSS 13.0统计软件及UNPHASE遗传分析软件进行统计分析。结果:未发现5-HTTLPR位点与吸烟行为的不同阶段包括吸烟启动(P=0.922)、吸烟维持(P=0.309)和尼古丁依赖(P=0.701)存在关联。进一步分层分析发现:携带S等位基因(SS+SL)的吸烟者中神经质人格特质与FTND总分正相关(r=0.118,P=0.004),与吸烟严重指数(HSI)正相关(r=0.102,P=0.013),与平均每天的吸烟量正相关(r=0.108,P=0.008),与第一次吸烟的年龄负相关(r=-0.117,P=0.004)。结论:5-HTT基因可能通过调节神经质人格特质与吸烟行为之间的关系而间接影响吸烟行为。

ASSOCIATION OF POLYMORPHISM IN THE SEROTONIN TRANSPORTER PROMOTE GENE WITH SMOKING BEHAVIORS IN CHINESE MALES

Objective:To explore the relationship between polymorphism in serotonin transporter gene(5-HTTLPR)and smoking behaviors in Chinese male lifetime nonsmokers and smokers.Methods:A total of 1012 subjects were assessed using smoking related questionnaire,Eysenck Personality Questionnaire(EPQ)and the Fagerstrom Test of Nicotine Dependence Questionaire(FTND).5-HTTLPR was investigated by polymerase chain reaction fragment length polymorphism techniques.The alleles and genotypes analysis were conducted with the UNPHASE program(version 3.0).Results:There were no significant association of 5-HTTLPR with smoking initiation(P=0.922),smoking maintenance(P=0.309)and nicotine dependence(P=0.701).Stratification analysis showed that neuroticism trait was positive related with FTND score(r=0.118,P=0.004),the heaviness of smoking index(HIS)(r=0.102,P=0.013),smoking quantity(SQ)(r=0.108,P=0.008)were negative related with first age of smoking(r=-0.117,P=0.004)in individual who carried S allele.Conclusion:Our data suggests that 5-HTTLPR gene may have no direct association with smoking behaviors,but it may affect smoking behaviors through modifying the effects of neuroticism.
