冷冻瘤苗对肿瘤浸润性淋巴细胞增殖及抗肿瘤活性的影响

目的研究冷冻瘤苗对肿瘤浸润性淋巴细胞(TIL)在体外增殖及抗肿瘤活性的影响。方法取人肝癌标本23例分离TIL,实验1组常规培养,实验2组加冷冻瘤苗进行培养,10天计数一次,每10天用3H-TdR释放法检测两组TIL杀伤自体肿瘤细胞及K562细胞的活性。结果实验1组40天后增殖力下降,实验2组保持高增殖力,两组细胞数有显著差异(P<0.05)。两组显示出高杀伤自体肿瘤细胞活性,与杀伤K562细胞相比有显著差异(P<0.01)。实验1组TIL20天后杀伤自体肿瘤细胞活性下降,实验2组仍保持较高的杀伤活性,两组差别有显著意义(P<0.05)。结论冷冻瘤苗能使TIL保持较长时间的增殖力,增强杀伤自体肿瘤细胞的活性。

Influence of frozen cancer vaccines on the proliferation and antitumor activity of tumor-infiltrating lymphocytes

Objective To study the influence of frozen cancer vaccines on the proliferation and antitu-mor activity of tumor-infiltrating lymphocytes ( TIL ) in vitro. Methods TILs were separated from23human liver cancer specimens.Experimental group1TIL was cultured with routine methods ,experimental group2TIL was cultured with frozen cancer vaccines.TILs were counted every10days ,and effect of killing autotumor cells and K562cells of TIL were tested with3H-TdR releasing method every10days. Results Proliferation of the TIL of experimental group1was reduced after40days.However ,proliferation of the TIL of experimental group2was kept in high level.The difference of cell quantity between two groups was significantly (P <0.05 ) .High activity of killing autotumor cells were displayed in both groups ,and there was significant difference compared to that of killing K562cells (P <0.01 ) .The activity of killing autotumor cells of experimental group1reduced after20days.However ,experimental group2was kept in high level of killing.The difference of antitumor activity between two groups was significantly (P <0.05 ) . Conclusion Frozen cancer vaccines can keep the proliferation of TIL and en-hance the activity of killing autotumor cell in vitro.
