电针对缺氧缺血性脑损伤幼鼠大脑皮质血流的影响

摘    要:目的 通过建立新生大鼠缺氧缺血性脑损伤(HIBD)模型,并检测电针干预治疗前后大鼠局部脑血流量(rCBF)的变化,开展针刺治疗神经系统疾病机制的研究.方法新生SD大鼠32只,随机分为假手术对照组、假手术+电针组、HIBD对照组和HIBD+电针组,每组8只.HIBD组予左侧颈总动脉结扎,休息2 h后置于温暖湿润的低氧环境处理2 h;假手术组不予结扎动脉及低氧处理.假手术+电针组和HIBD+电针组大鼠于造模次日开始予电针百会、大椎穴,30 min·d-1,14 d为1个疗程.假手术对照组、HIBD对照组仅固定四肢,不予针刺.分别在针刺疗程的第1天、第7天、第14天对假手术+电针组、HIBD+电针组幼鼠进行皮质脑血流监测(n=4),并于疗程结束后次日(日龄22 d)进行运动功能评价.结果疗程的早、中、晚期,电针假手术组或脑损伤组大鼠均可致rCBF增强;与假手术对照组比较,HIBD对照组的运动功能(悬吊试验和斜坡试验)受损严重;HIBD+电针组大鼠的运动功能较HIBD对照组明显恢复.结论电针可促进脑损伤大鼠皮质的rCBF,有助于脑损伤和运动功能的恢复.

Abstract:Objective To explore the machanism which how elecrtro-acupuncture(EA) affects hypoxic-ischemic brain damage(HIBD) rats,via the monitoring of regional cerebral blood flow(rCBF).Methods Thirty-two healthy Sprague-Dawley neonatal rats were divided into sham control group(n=8),sham+EA group(n=8),HIBD control group(n=8) and HIBD+EA group(n=8).HIBD rat models were established on their 7 days old.Briefly,the left carotid artery was isolated and ligated.Rats were returned to their mothers for a 2-hour recovery,and then they were placed into a glass jar for 2 hours of hypoxic environment.In sham rats,the left carotid artery was exposed and isolated without ligation and was not treated.From the next day,rats in sham+EA group and HIBD+EA group were electric stimulated 30 min daily for 14 days,with BAIHUI and DAZHUI as the acupoints.Rats in sham control and HIBD control groups were just fixed,without acupuncture at the same time.rCBF was monitored before,during and after EA at different periods of intervention(the 1st day,7thday and 14thday sessions) in rats undergoing EA(n=4).Motor function was assessed on the 2nd day of the treatment course.Results In the early,middle or late stages of the treatment course,EA would improve rCBF of rats in HIBD and sham groups.Compared with sham control group,motor function of HIBD control group was seriously impaired.But EA improved the damaged motor function.Conclusion EA can improve motor function of HIBD rats,through its improvement of rCBF.
