改良Bouvet法纯化IgM型单克隆抗体

摘    要:利用IgM的优球蛋白特性,对Bouvet法进行了改良。比较了Bouvet法和改良法对两种IgM型McAb腹水的纯化。结果表明,改良法保留了Bouvet法的优点,而荷载量大大高于Bouvet法,获得了良好的抗体纯度和回收率(1.0~1.5mg/ml腹水)。说明此法为一种简单、快速、高效的纯化IgM型单克隆抗体的方法。

Abstract:Based on the euglobin characteristic of IgM, a modified method in which euglobin precipitation and Bouvet method were combined was established. Two IgM momoclonal antibodies (McAb) A12 and PC 1 were purified with the modified method in comparasion with Bouvet method. It was shown that the modified method has a simplicity as does Bouvet method. Moreover, the sample loading amount in the modified method was higher than Bouvet method by 5-10 times, and the yield could reach 1.0-1.5mg/ml. Therefore, the modified method could be used as routine to purify IgM McAb with simplicity, high efficience and more yield.
