大分子葡萄糖酐血瘀证动物模型表征的偶氮蓝变化特征

摘    要:目的探讨偶氮蓝与不同浓度大分子葡萄糖酐所致血瘀证动物模型血瘀程度表征变化规律的关系。方法用0.5%偶氮蓝配制的5%、10%、15%的大分子葡萄糖酐造模,观察偶氮蓝在大分子葡萄糖酐所致血瘀证模型动物耳廓、舌体呈现出的体表表征变化,检测耳廓、血浆中偶氮蓝含量。结果糖酐各组1h后耳廓及舌体出现不同程度蓝色瘀斑,且与大分子葡萄糖酐注射浓度呈正相关。同对照组大鼠相比,糖酐各组大鼠1h后耳廓偶氮蓝含量随大分子葡萄糖酐注射浓度的增加而增加,血浆偶氮蓝含量则随其注射浓度的增加而减少。结论大鼠尾静脉注射用0.5%偶氮蓝配制的不同浓度的大分子葡萄糖酐后能使血瘀证模型大鼠的血瘀程度在其耳和舌体等体表表征中较为直观地呈现出来,且大分子葡萄糖酐注射浓度不同所致血瘀证模型大鼠耳廓和血浆偶氮蓝含量变化与耳廓、舌体在同一时间出现的血瘀程度明显相关,说明偶氮蓝可以客观反映大分子葡萄糖酐所致血瘀证模型大鼠血瘀程度的表征变化。

Abstract:Objective To discuss the relationship between Evans blue and the changing rules of exosymptoms of animal model with blood-stasis syndrome induced by macromolecular dextran in different concentrations.Method The rat model was established by using different concentrations of macromolecular dextran (5%, 10% and 15% compounded with 0.5% Evans blue).The exosymptom changes presented by Evans blue in ear and tongue body of rat model with blood-stasis syndrome induced by macromolecular dextran were observed and the contents of Evans blue in the ear and plasma were detected.Result After one hour there were blue ecchymosis in different degrees in model ear and tongue body in different groups injected different concentrations of macromolecular dextran.The ecchymosis was correlated with the concentration of macromolecular dextran.Comparing to the rats in the control group the rats in different macromolecular dextran groups had a higher content of Evans blue in ear with a higher concentration of macromolecular dextran, while the content of plasma Evans blue decreased when the concentration of macromolecular dextran increased.Conclusion The model rats injected in caudal vein with different concentrations of macromolecular dextran (compounded with 0.5% Evans blue) can present the exosymptoms in the ear and tongue body directly.The content changes of plasma Evans blue in the ear and tongue body are correlated with the degree of blood stasis at the same time, which indicates that Evans blue can reflect objectively the exosymptom changes in rat model of blood-stasis syndrome induced by macromolecular dextran.
