| 摘 要： | 目的:探索黄芪含药血清诱导骨髓间充质干细胞(MSC)分化为心肌样细胞的作用。方法:采用日本大耳白兔制备黄芪含药血清,分离SD大鼠MSC进行传代培养。用黄芪含药血清和5-Aza进行诱导分化,免疫组化法检测诱导后细胞心肌肌钙蛋白、结蛋白表达,RT-PCR反应测定α-MHC和β-MHC表达。结果:诱导后细胞心肌肌钙蛋白、结蛋白表达阳性,并可以表达心肌特异性的肌球蛋白。结论:黄芪含药血清可诱导MSC分化为心肌样细胞。

| Abstract: | Objective:Study on the effection of marrow mesenchyma stem cell differentiate into the cardiac muscle type cell inducted by the medicine blood serum with Huangqi.Method:Uses the Japanese big ear white rabbit to prepare the medicine blood serum with Huangqi.Separates SD rat MSC,cultivate and passage it.Then induct and differentiate it with the medicine blood serum and 5-Aza.Detect the protein expression of the inducted cell by immunohistochemical method.Detect the expression of myoglobulin by RT-PCR method.Result:The protein of cTnT and Desmin express positive in the inducted cell and express the specificity myoglobulin of cardiac muscle.Conclusion:The medicine blood serum with Huangqi may induce MSC to differentiated into the cardiac muscle type cell. |
