P16和nm23-H1在乳腺癌组织中的表达及其意义

摘    要:目的探讨新型抑癌基因P16和肿瘤转移抑制基因nm23-H1在乳腺癌中的表达及其与临床病理因素的相关性。方法应用免疫组织化学SP法检测50例乳腺癌手术标本和25例乳腺纤维腺瘤标本(阴性对照组)中P16和nm23-H1的表达,通过光学显微镜观察免疫组织化学表达情况并应用统计学方法对结果进行分析。结果P16的阳性表达率在乳腺癌组明显低于对照组(P〈0.05)。nm23-H1的阳性表达率在阳性组和对照组间比较无显著性差异(P〉0.05)。在乳腺癌中,nm23-H1的阳性率在各临床分期组间比较有显著性差异(P〈0.01);P16和nm23-H1的阳性率在各组织学分级组间比较有显著性差异(P〈0.01);nm23-H1和P16的阳性表达率在有无淋巴结转移组之间比较有显著性差异(P〈0.05)。结论联合检测乳腺癌组织中P16和nm23-H1的表达状况有助于判断预后。

Abstract:Objective To study the expression of growth suppressor gene P16 and nm23 - H1 and their correlation with clinical pathological factors in breast cancer. Methods S - P method of Immunohistochemistry was applied to detect the expressions of P16 and nm23 - H1 in 25 cases of breast fibroadenoma and in 50 cases of breast cancer. The staining result was observed by optical microscope. Results The positive expression rate of P16 (48%) in breast cancer was statistically lower than that in fibroadenoma(88% ) . while nm23 - H1 (58%) was also lower than that in fibroadenoma(72% ) and showed no significance (P 〉0.05 ). The positive expression rate of P16 and nm23 - H1 were significantly associated with histological grading ( P 〈 0.01 ). The positive expression rate of P16 and nm23 - H1 were significantly associated between with or without lymph node metastasis ( P 〈 0.05). The expression of nm23 - H1 was significantly associated with TNM stage ( P 〈 0.01 ). Conclusion It could help to judge patients'prognosis by jointly detecting the expression rate of P16 and nm23 - H1 in breast cancer
