人胃癌移植瘤裸小鼠Ki-ras基因突变的检测

摘    要:目的:检测人胃癌裸小鼠移植瘤Ki—ras基因的突变情况。方法将人胃癌细胞SGC-7901于裸小鼠脾包膜下移植,12周后处死,采用PCR-DNA测序法检测Ki—ras基因第12、13密码子的突变情况。结果:共检测29例裸鼠移植瘤,DNA经扩增后20例为阳性,其中12例进行DNA测序,未发现第12、13密码子的突变。结论:人胃癌移植瘤裸小鼠Ki—ras基因第12、13密码子有无突变尚不能肯定。

Abstract:Objective To detect the mutation of Ki ras gene exon 12, 13 of human gastric cancer xenografts in nude mice. Methods A human gastric adenocarcinoma cell line SGC 7901 was grafted under splenic capsule of 32 mice, Ki ras gene exon 12, 13 were detected by PCR DNA method 12 weeks after the procedure. Results No Ki ras gene exon 12, 13 mutations were detected. Conclusions The presence of mutation of Ki ras gene exon 12, 13 on human gastric cancer xenografts of nude mice can not be confirmed.
