摘    要:用高效凝胶渗透色谱法(HPGPC)鉴定了茶叶糖蛋白(TGC)的纯度,并以TGC纯品作为标准品,以UltrahydrogelTM 500(7.8×300 mm)为固定相,0.6 mL/min蒸馏水为流动相,折光示差检测,对多种茶叶中TGC含量进行了测定,得回归方程为:Y=0.250 3×10-6-0.008 7,r=0.999 7,其浓度在0.604~6.04mg·mL-1范围内呈良好的线性关系。

Abstract:The purity of tea glycoprotein (TGC) was determined by high performance gel permeation chromatography (HPGPC). Then, this TGC was used as the standard substance for the determination of TGCs in various teas. The chromatographic conditions were with UltrahydrogelTM 500(7. 8×300 mm) column as stationary phase and the distilled water as the mobile phase at a flow rate of 0. 6 mL·min 1 at 35 C and the refractive index (RI) detector. The calibration curve was linear over the concentration range of 0. 604-6. 04 mg·mL-1 for TGC. The correlation coefficient was r = 0. 9997. The determination method of TGC developed in this paper was found to be simple, accurate and reliable.
