摘 要: | 目的观察骨髓间充质干细胞(BMSCs)对博来霉素致小鼠肺损伤动物模型的疗效。方法体外分离、培养雄性6周C57BL/6J小鼠间充质干细胞。15只雌性8周C57BL/6J小鼠随机分为3组:阴性对照组(P组),阳性对照组(B组),BMSCs治疗组(B+M组,每组5只)。P组于气管内注入PBS 50μL,B组及B+M组按5μg/g剂量气管内注入博莱霉素50μL制备肺纤维化模型,B+M组于造模后6 h经尾静脉注射BMSCs 1×107/mL,200μL。实验第28天统一处死小鼠,留取肺组织行病理HE染色、羟脯氨酸(hydroxyproline,HYP)含量测定,提取肺组织的DNA,通过聚合酶链反应(PCR)-琼脂糖电泳法检测雄鼠的性别决定基因(sry基因),以判断外源给予的BMSCs是否在雌性小鼠肺组织中存在。结果①肺组织病理:B+M组的肺泡炎及肺纤维化程度均较B组减轻。②HYP含量:B组的HYP含量明显高于P组及B+M组,B+M组的HYP含量与P组无明显差异。③B+M组有雄性小鼠的sry基因存在。结论①外源性BMSCs经尾静脉注射可以到达肺损伤部位;②外源性BMSCs可以减轻肺泡炎、肺纤维化的程度。
