摘    要:目的:观察表皮细胞生长因子(BGF)、碱性成纤维生长因子(bFGF)体外诱导骨髓间充质干细胞(MSCs)分化为神经细胞后,神经元微管相关蛋白Tau和神经元特异性烯醇化酶(NSE)的表达。方法:取P4代MSCs分为EGF、bFGF和EGF+bFGF及对照组;免疫细胞化学法检测Tau蛋白和NSE表达,ELISA法分析各组细胞Tau蛋白含量。结果:免疫细胞化学检测显示EGF+bFGF811Tau蛋白和神经元特异性烯醇化酶NSE阳性细胞率最高,bFGF组、EGF组和对照组依序递减;各组Tau蛋白含量结果与Tau蛋白阳性细胞率一致。结论:EGF和bFGF均可促进MSCs向神经细胞分化,并表达Tau蛋白和NSE,二者具有协同作用。

Abstract:Objective: To observe the expression of Tau protein and neuron-specific enolase (NSE) after differentiation of bone-marrow mesenchymal stem cells (MSCs) into neural cells induced by EGF and bFGF in vitro. Methods: P4-MSCs were divided into 4 groups: EGF+bFGF, bFGF, EGF, and control groups. After 14-day induction, positive rates of Tau protein and NSE were tested by immunocytochemistry and Tau protein content assay was analyzed with ELISA test. Results : It was found that the positive rates of Tau protein and NSE in EGF+bFGF group were the highest, followed by bFGF, EGF and control groups in turn (P<0. 001). In eachgroup, the content of Tau protein was in consistent with its positive rate. (P<0. 001). Conclusion: EGF and bFGF can induce MSCs into neural cells in vitro and express Tau protein and NSE. They have synergistic effects during the inducing process.
