保肝Ⅰ号对急慢性肝损伤的保护作用

摘    要:目的:探讨傣药保肝Ⅰ号(BGYH)对急慢性肝损伤的保护作用.方法:采用iv卡介苗(BCG,1.5 ×107个活菌数/只)+脂多糖(LPS,10μg/只)造成小鼠急性免疫性肝损伤模型,后连续ig保肝Ⅰ号低、中、高(5.1,10.2,20.4 g·kg -1)剂量12 d;采用iv刀豆蛋白(Con A,15 mg·kg-1)造成小鼠急性免疫性肝损伤模型,后连续ig保肝Ⅰ号低、中、高(5.1,10.2,20.4g· kg-1)剂量7d;采用sc CC14(40%)大豆油溶液建立大鼠慢性肝纤维化模型,后连续ig保肝Ⅰ号低、中、高(0.14,0.28,0.56g·kg-1)剂量8周.各组动物取血,测定血清谷丙转氨酶(ALT)和谷草转氨酶(AST)活力,并取肝脏做病理切片,观察肝组织病变,测定血清肿瘤坏死因子-α(TNF-o),白蛋白(Alb),测肝组织中羟脯氨酸(Hyp)含量.结果:在BCG+ LPS模型中,保肝Ⅰ号能显著降低ALT,AST活力和肝脏指数(HI);在ConA模型中,保肝Ⅰ号能显著降低AST,ALT,TNF-α的活力;在CC14复合因素诱导的大鼠慢性肝损伤模型中,保肝Ⅰ号能显著降低AST,ALT,升高Hyp,能显著升高Alb含量,且保肝Ⅰ号能减轻各模型动物肝脏病变.结论:保肝Ⅰ号对急慢性肝损伤具有较好的保护作用.

Abstract:Objective: To investigate the protective effect of Dai medicine Baogan Yihao (BGYH) on acute and chronic liver injury. Method: Three kinds of animal model were established in this study. Acute model of immunological hepatic injury was established by iv Bacillus Calmetle-Guerin(BCG)+Lipopolysaccharide(LPS) in mouse, and ConA respectively. Rat model of chronic hepatic fibrosis was developed by ih of (CCl4). BGYH with high, medium and low doses were orally administrated. Activity of alanine aminotransferase (ALT) and spartate aminotransferase (AST) in serum was determined, and pathological changes in liver were also examined. Result: BGYH at the concentration of 20.4 g·kg-1 showed significantly inhibition on the acute immunological liver injury in mice induced by BCG+LPS and ConA; BGYH at the concentration of 0.56 g·kg-1 also inhibited the chronic hepatic fibrosis induced by CCl4 in rat model. And BGYH showed protection for hepatic lesions in animal models. Conclusion: BGYH can obviously protect acute and chronic liver injuries.
