钒酸盐对实验性糖尿病大鼠肾脏的影响

摘    要:目的 观察钒酸盐(Va)对链脲霉素诱导的糖尿病(DM)大鼠肾脏功能和结构的影响。方法 正常大鼠和糖尿病大鼠自由饮用钒酸钠水12周后,包括内生肌酐清除率、脲蛋白排泄量(UPE),并取肾脏作光镜和电镜检查,测定肾小球平均面积(MGPA)和肾小球平均体积(MGV)。结果DM大鼠12周时存在肾功能改变,Ccr和UPE增加,MGPA和MGV增大,并有肾脏超微结构受损,而钒酸盐治疗能明显改善DM大鼠的上述改变

Abstract:Objective: To study the effects of Vanadate(Va) on renal function,and structure in streptozotocin induced diabetic rats and to provide the experimental bases for clinical application of Va. Methods: Sodium orthovasnadate was administered to normal and diabetic rats. 24h urinary protein excretion(UPE) and clearance of creatinine(Ccr) were determined in normal rats and diabetic rats at 12nd week,and the kidaneys were processed for morphological examination with light and eletron microscopes. Mean glomerular plane area (MGPA) and mean glomerular volume(MGV) were calculated. Results: Ccr.LKW.MGPA .and MGV increased at 12nd week in diabetic rats. The electron microscopic results showed that the ultrastructures of kidney were significanty deranged in diabetic rats. Ccr.LKW.MGP and MGV in Vatreated diabetic rats decreased to approach normal level,and the pathologic changes of kidney were improved significantly. In Va-treated noraml rats,no changes were found in renal function. No increase of plasma insulin was found. Conclusion: Va could contribute to the improvement of diabetic nephropathy in diabetic rats.
