犬外周血基质细胞成骨诱导分化后的超微结构观察

摘    要:目的:利用透射电子显微镜(Transmission electron microscope,TEM)观察成骨诱导培养前后,犬外周血基质细胞(dog peripheral blood stromal cells,dPBSCs)超微结构的变化。方法:采用密度梯度离心法—贴壁筛选法分离培养dPBSCs,并进行成骨诱导培养,14d后进行TEM观察。结果:经成骨诱导培养的dPBSCs光镜观察显示,形态由长梭形变为短梭形成骨细胞样形态,体积增大,细胞排列较杂乱;透射电镜观察显示,细胞体积增大,胞质中含有丰富的细胞器,还有大量的基质小泡、呈同心圆状或板层状的髓样体和空泡状结构,细胞间连接紧密,带有大量微绒毛样突起。结论:dPBSCs是一种具有成骨潜能的细胞,经成骨诱导培养后,其超微结构具有成骨细胞的特点。

Abstract:Objective:To observe the ultrastructure of dog peripheral blood stromal cells(dPBSCs)after the osteoinductive culture and non-induced culture with a transmission electron microscope.Methods: PBSCs were isolated from the peripheral blood of dog via density gradient centrifugation and cultured in the osteoinductive medium with 10nmol/L dexam-ethasone,0.1mmol/L ascorbate and 10mmol/L β-glycerol phosphate or in ordinary culture for 14 days.Transmission electron microscopy was used to observe the ultrastructural changes.Results: After osteoinductive culture,light microscopy showed that the cells became bigger;TEM showed that there were richer mitochondria,endoplasmic reticulum and Golgi complex in the cytoplasm of dPBSCs than that in the ordinary clutured dPBSCs.And lots of myeloid like structures and matrix vesicles with middle or low electron density were observed in the dPBSCs cultured with the osteoinductive medium.Conclusion: The ultrastructural characteristics of dPBSCs after the osteoinductive culture were similar to those of osteoblasts.
