布地奈德对哮喘模型大鼠肺组织Toll样受体4和5表达的影响

摘    要:目的观察布地奈德对哮喘模型大鼠肺组织Toll样受体4(TLR4)和TLR5表达的影响,探讨TLR在哮喘炎症机制中的作用。方法 27只SD大鼠随机分成哮喘组、对照组、布地奈德组,每组9只。制备哮喘大鼠模型,免疫组织化学法检测各组大鼠肺组织TLR4和TLR5表达。结果哮喘组大鼠肺组织TLR4光密度值与对照组比较,差异无统计学意义(P〈;0.05);布地奈德组大鼠肺组织TLR4光密度值显著低于对照组和哮喘组(P〈0.05)。哮喘组大鼠肺组织TLR5光密度值显著高于对照组(P〈0.05);布地奈德组TLR5与对照组及哮喘组比较,差异均无统计学意义(P〈0.05)。肺组织TLR4和TLR5蛋白表达水平无相关性(r=0.246,P>0.05)。结论 OVA致敏的哮喘模型大鼠肺组织TLR5表达升高,TLR4无变化,TLR5在哮喘中可能起到促炎作用。布地奈德能下调哮喘模型大鼠肺组织TLR4和TLR5表达。

Effect of Budesonide on the Expression of Toll-like Receptor 4 and 5 in Rat Asthma Model

Abstract:Objective To investigate the effect of budesonide on the expression of Toll-like receptor (TLR) 4 and 5 in asthma rats and the potential roles of TLR in pathogenesis of asthma inflammation. Methods Twenty-seven SD rats were randomly divided into three groups:asthma group,control group,and budesonide group. The rat model of asthma was established with injection of OVA. The expressions of TLR4 and TLR5 proteins were detected by immunohistochemical method. Results No significant difference in TLR4 expression in lung tissue was noted between asthma group and control group (P〉0.05). The level of TLR 4 in budesonide group was significantly lower than that in control group and asthma group(Ps〈0.05). The expression of TLR5 in lung tissue in asthma group was dramatically higher than that in control group(P〈0.05),while it did not differ between budesonide group and control group or asthma group (Ps〉0.05). No correlation between the levels of TLR4 and TLR5 in lung tissue was found (r=0.246,P〉0.05). Conclusion In OVA challenged asthmatic lung tissue,the level of TLR5 was elevated,while the level of TLR4 did not change,indicating that TLR5 may be as a inflammation-promoting mediator in asthma exac-erbation. Budesonide can down-regulate the levels of TLR4 and TLR5.
