血小板源性生长因子促进新生血管形成的机制研究

摘    要:目的 :观察血小板源性生长因子 ( PDGF)对于培养脐静脉内皮细胞碱性成纤维细胞生长因子 ( b FGF)水平的影响 ,揭示 PDGF促进新生血管形成的机制。方法 :免疫组织化学检测培养脐静脉内皮细胞 b FGF水平。结果 :缺氧培养与常规培养相比内皮细胞胞浆b F GF水平无显著变化 ;不同剂量的 PDGF在常规或缺氧培养条件下均能增加内皮细胞胞浆 b FGF水平 ,且呈剂量依赖性。结论 :说明 PDGF可能通过直接促血管形成生长因子的介导作用间接促进新生血管的形成。

Abstract:Objective: To investigate whether platelet derived growth factor(PDGF) known to promote neovascularization in vivo could modulate the expression of basic fibroblast growth factor(bFGF) in human umbilical endothelial cells. Methods: The level of bFGF of endothelial cell was detected with Immunhistochemical. Results: The level of bFGF had no changes when cell under hypoxia condition. Various contents of PDGF doses dependently induced bFGF synthesis under normal or hypoxia. Conclusion: These manifest PDGF promoted angiogensis by increasing bFGF levels of endothelial cells.
