摘    要:目的探讨他莫昔芬(Tamoxifen,TAM)对人子宫内膜癌细胞Ishikawa细胞周期及细胞内Ca^2+的影响。方法利用四甲基偶氮唑蓝(MTT)比色法测定TAM对细胞增殖量效的影响;采用流式细胞仪检测TAM对Ishikawa细胞周期的影响;应用激光共聚焦显微镜动态检测TAM作用后细胞内Ca^2+的变化。结果TAM浓度为1×10^-9~1×10^-6mol/L时,呈剂量依赖性促进Ishikawa细胞的增殖,TAM浓度为1×10^-7mol/L时,作用最显著;TAM(10^-7mol/L)可促进细胞周期由G0/G1期进入S期,G0/G1期的DNA含量由51.58%下降到44.85%,S期的DNA含量由34.73%上升到43.64%;TAM使细胞内钙的荧光强度快速升高,约在20 s后开始增高,90 s左右到达峰值,约为静息状态的2.52倍,持续约120 s。结论TAM通过调节细胞周期和介导细胞内Ca2+浓度的快速增高来促进Ishikawa细胞增殖。

Abstract:Objective To explore the effect of tamoxifen on the intracellular Ca2+ level and cell phases of human endometrial carcinoma Ishikawa cells.Methods MTT assay was used to determine the cell proliferation after TAM treatment.Change of cell phase induced by TAM was determined by Flow-cytometry.Laser scanning confocal microscope was used to continuously monitor the change of intracellular free calcium induced by TAM.Results At the concentration of 1×10^-9~1×10^-6mol/L,Tamoxifen stimulated the proliferation of Ishikawa cells in a dose-dependent manner especially at the concentration of 1×10^-7mol/L.TAM Stimulated the proliferation of Ishikawa cells at G1/S phases.It blocked G1 phase from 51.58% to 44.85%,and stimulated S phase from 34.73% to 43.64%.After TAM treatment,the intracellular Ca2+ level increased rapidly at the 20 second and reached the peak level at the second,which was 2.5 times of the static status cells.It maintained for about 140 seconds.Conclusions TAM Stimulated the proliferation of Ishikawa cells at G1/S phases.TAM can Stimulate the release of the intracellular Ca^2+ in a rapid manner.
