川芎嗪对大鼠骨髓间充质干细胞长期诱导效应的研究

目的 探讨川芎嗪提取液体对大鼠骨髓间充质干细胞(MSCs)分化为神经元样细胞的长期诱导效应.方法 分离培养大鼠骨髓MSCs,用1.50g/L川芎嗪提取液对MSCs进行诱导,倒置显微镜下观察诱导培养不同时间后细胞的形态变化,并应用免疫细胞化学方法鉴定诱导培养后细胞浆内Nestin和NSE蛋白表达.结果 川芎嗪诱导培养8h后即可见细胞形态发生变化,24 h后表现为典型的神经元细胞形态,Nestin和NSE神经细胞特异性标志物呈阳性表达,3d后诱导效果最好,6d后,有些细胞突起变长,出现分叉,并且有些细胞出现衰老死亡.结论 川芎嗪早期诱导BMSCs向神经元样细胞分化的效果较好,长期诱导的特异性不明显.但可促进细胞突起的生长,并且促进神经细胞的成熟和死亡.

Research of Ligustrazine Induced Effects of Long-term to Rat Bone Marrow of Mesenchymal Stem Cells

Objective To investigate the effects of Long-term that ligustrazine induced differentiation of rat bone marrow mesenchymal stem cells (BMSC) into neuron-like cells. Methods BMSC from rat were isolated and cultured. Then BMSC were induced by ligustrazine (1.50g/L), the morphological changes of the cells were observed under inverted microscope. The neuronal specific markers: Nestin and neuron specific enolase (NSE) were detected by immunocytochemistry staining. Results After induction for 8h, some MSCs showed morphological changes. Twenty-four hours later, the differentiated cells showed typical neuronal morphology, and expressed neuron-specific markers, such as Nestin and NSE, the best effects of inducing after three days, six days later, some cells bumps getting longer and branching,and some cells appear aging and death. Conclusion Ligustrazine induces early the differentiation of BMSC into neuron-like cells in better effect, the specificity of the long-term induction is not obvious.But it can promote the growth of cells bumps,and promote mature and death of nerve cells.
