摘    要:目的:初步探讨超氧化物歧化酶(SOD)、细胞内活性氧(ROS)在三氧化二砷(As2O3)抗肿瘤中的作用。方法:对小鼠S180肉瘤细胞采用细胞培养,并建立S180移植瘤动物模型,体内体外观察As2O3对S180肉瘤的抑制作用,同时检测S180肉瘤细胞及S180移植瘤肿瘤组织内的总超氧化物歧化酶(TSOD),含锰超氧化物歧化酶(MnSOD),含铜锌超氧化物歧化酶(CuZnSOD)以及ROS的改变。结果:As2O3可以抑制小鼠S100肉瘤细胞生长并具有浓度依赖性,在一定浓度下其作用强于化疗药物阿霉素(ADM);肿瘤细胞内及肿瘤组织内的TSOD以及MnSOD均表现出下降的趋势,ROS则表现出上升趋势。TS0D和ROS呈负相关。结论:As2O3是通过降低细胞内SOD值,使细胞内超氧阴离子(O2^*-)更多的积累以杀伤肿瘤细胞,这可能是As2O3,抗肿瘤作用机制之一。

Abstract:Objective: To investigate the roles of arsenic trioxide in antitumor process associated with superoxide dismutase. Methods:S180 cell and tumor-bearing mice were treated in different concentrations of As2O3, then the inhibiting effect on tumor by MTT detected and the TSOD,CuZnSOD,MnSOD,ROS in tumor cell and tumor tissue observed. Results: The S180 cell was inhibited not only in vito but also in vitro, with the increasing concentration of As2O3. The TSOD,MnSOD,CuZnSOD were decreased, the reactive oxygen species(ROS) were increased. Significant relationships were found between TSOD and ROS. Conclusion:SOD plays an important role in the antitumor process of As2O3.
