摘    要:目的探讨血管内皮生长因子(VEGF)对软骨细胞凋亡的影响。方法乳鼠关节软骨细胞体外培养成功后,实验分为三组。每组加入不同处理因素进行干预,对照组:不加任何处理因素;VEGF组:VEGF 10 ng/m L;白介素(IL)-1β组:IL-1β10 ng/m L,连续培养48 h。采用流式细胞仪检测软骨细胞的凋亡率,采用蛋白免疫印迹法(Western Blot)检测增殖细胞核抗原(PCNA)蛋白的表达,通过硝酸还原酶法检测上清液中一氧化氮(NO)的含量。结果软骨细胞的凋亡率VEGF组(9.28±2.35)%]及IL-1β组(17.14±5.07)%]明显高于对照组(2.83±0.77)%];软骨细胞PCNA蛋白表达VEGF组(0.86±0.24)及C组(0.72±0.05)明显低于对照组(1.01±0.11);软骨细胞分泌的NO含量VEGF组(112.31±12.73)μmol/L]及IL-1β组(150.02±15.34)μmol/L]显著高于对照组(49.35±6.68)μmol/L],差异均有高度统计学意义(P〈0.01)。结论 VEGF能够介导软骨细胞凋亡,抑制软骨细胞的增殖活性。

Effects of vascular endothelial growth factor on apoptosis of chondrocytes

Abstract:Objective To investigate the effects of vascular endothelial growth factor(VEGF) on apoptosis of chondrocytes. Methods Chondrocytes were isolated and cultured, then chondrocytes were divided into 3 groups. Control group:without any disposal; VEGF group: chondrocytes were treated with 10 ng/m L VEGF; IL-1β group: chondrocytes were treated with 10 ng/m L IL-1β. After 48 h, the apoptosis rate of chondrocytes was detected by flow cytometry, the protein expression of PCNA was detected by Western Blot, and the content of nitric oxide in the supernatant was examined by nitrate reductase kit. Results The apoptosis rate of chondrocytes in VEGF group (9.28 ±2.35)% ] and IL-1β group(17.14±5.07)%] were significantly increased than that in control group (2.83±0.77)%], the expression level of PNCA protein in VEGF group(0.86±0.24) and IL-1β group(0.72±0.05) were less than that in control group(1.01±0.11). The content of nitric oxide in VEGF group (112.31±12.73) μmol/L] and IL-1β group (150.02±15.34) μmol/L] were significantly increased than that of control group (49.35±6.68) μmol/L], the difference was statistically significant(P〈0.01).Conclusion VEGF can induce the apoptosis of chondrocytes and inhibit the proliferation activity of the chondrocytes.
