并殖吸虫McAb识别靶抗原的IFAT固相抗原定位研究

摘    要:本文报告应用16 种单克隆抗体(McAb)以间接荧光抗体试验(IFAT),对卫氏并殖吸虫(P.w)、斯氏狸殖吸虫(P.s)及三平正并殖吸虫(E.c)成虫及其子宫部位虫卵的冰冻切片作靶抗原的定位研究证明:以P.w成虫抗原制备的杂交瘤分泌的8种McAb中,4种具有高度特异性,只识别P.w的有关抗原,另4种与P.s或E.C出现交叉反应;以P.w囊蚴抗原所制备的杂瘤分泌的8种McAb中,2种具有高度特异性,只识别P.w的有关抗原,1种只识别E.c抗原,1种与E.c出现交叉反应,另4种不识别固相抗原片上的抗原.这些McAb识别抗原的部位主要为肠上皮、皮层、食管、排泄囊及卵壳.识别P.w 抗原显示的荧光均较P.s、E.c者为强.

Abstract:Indirect fluorescent antibody test (IFAT) was used to look for the Paragonimus target antigens recognized by 16 McAb to either metacercariae (PM) or adult worms (PA) on adult worm and egg frozen section of Paragonimus westermani (P. w) ,Pagumogonimus skrjabini (P. s) and Euparagonimus cenocopiosus (E. c).The results showed that 4 PA McAb only, recognized P. w antigen without any cross-reaction, therefore they were of high specificity,4 other PA McAb had cross-reaction with E. c and P. s. 2 PM McAb only recognized P. w antigen without any cross-reaction,therefore they had high specificity also.1 PM McAb only recognized E. c antigen,1 PM McAb had cross-reaction with E. c,4 other PM McAb did not recognize the antigen on frozen section at all.The target antigens mainly located in the cecum,tegument,esophagus,excretory bladder,and egg shell. P. w target antigens showed much intenser fluorescence than P. s and E. c target antigens.
