黄芪甲苷提取与测定方法的研究

 目的 研究黄芪甲替提取方法及HPLC检测条件。方法 色谱柱：Inertsil ODS-3(5μm,4.6 mm×250 mm)；流动相：乙腈-水(35：65)；流速：1.0 ml·min〈sup〉-1〈/sup〉；蒸发光散射检测器参数：蒸发温度：100°C,雾化温度：80°C,出口温度：60°C，载气流速：1.5ml·min〈sup〉-1〈/sup〉。结果 80%甲醇索氏提取4h，水饱和正丁醇萃取3次，0.15 mol·L·min〈sup〉-1〈/sup〉氢氧化钠或40 %氨水洗涤3次；黄芪甲苷线性范围为4.582～22.913，平均回收率为100.4%,RSD=4.27%。结论 本法简便、灵教、准确。

Study on extraction and HPLC method of astragaloside IV

OBJECTIVE To study the extraction method and the HPLC determination condigtions in order to determine the con-tent of astragaloside IV in Radix Astragals .METHODS With a Inertsil ODS3(5 pm) column (4.6 mmx×250 mm) and CH3CN-HZO(35:65) as mobile phase, HPLGELS was used to determine the Astragaloside mntent. ELS condition: evaporator 100，nebu-lizer exhaust 60 and 1 .5 ml·min^-1gas speed. RESULT MeOH(80%)extraction time was 4 h,n-BuOH extracted 3 times and washed with NaOH(0.15 mol·L^-1)or ammonia solution (40%)3 times. Linear range of Asstragaloside IV was 4. 582 22. 913 pg. The recovery rate was 100.4% (RSD=4.27%).CONCLUSION This method provides a simple and feasible way to determine astragaloside IV.
