浙江地区健康青少年和成人外周血T淋巴细胞免疫表型调查及正常参考值范围的建立

目的通过对健康青少年和成人外周血T淋巴细胞免疫表型分布调查，建立健康人外周血T淋巴细胞免疫表型正常参考值范围。方法选择浙江地区义务献血和健康体检者385例，血样用Multi TEST四色特异性T淋巴细胞亚群荧光抗体进行荧光标记，FACS Callibur流式细胞仪检测样品，Mulfiset程序获取数据并分析结果，SPSS 12．0软件包进行统计分析。以百分位数法，采用95％可信区间确定正常参考值范围。结果男性和女性的CD3^＋的百分率和CD3^＋CD4^＋百分率和绝对数均有差异，表现为女性高于男性。而CD3^＋绝对数和CD3^＋CD8^＋百分率和绝对数没有性别差异。T淋巴细胞免疫表型各个指标在不同年龄组之间均有差异，表现CD3^＋CD8^＋（相对数和绝对值）随年龄组增大而逐渐下降趋势，而CD3^＋CD4^＋（相对数和绝对值）则随年龄组增大而逐渐增加。浙江省健康人T淋巴细胞免疫表型参考值范围：CD3^＋百分数（％）：47．98～82．60；CD3^＋绝对数（个／μl）：712．49～2367．55；CD8^＋百分率（％）：14．24～41．48；CD8^＋绝对数（个／μl）：219．14～1109．81；CD4^＋百分率（％）：24．15～51．66；CD4^＋绝对数（个／μl）：383．61～1345．60；CD4／CD8：0．70～3．25。结论健康人淋巴细胞免疫表型的分布存在性别和年龄差别。由于这种差别细微，可以在同一种族同一区域建立统一的正常参考值范围。

Investigation on T Lymphocyte Subsets in Healthy Teenagers and Adults in Zhejiang Areas and Establishment of the Reference Range for Normal Values of T Lymphocyte Subsets

Objective To detect T lymphocyte subsets in healthy teenagers and adults in Zhejiang areas and establish the reference range for nomud values of T lymphocyte subsets. Methods 385 blood samples collected from blood donors and health examination were stained with four color fluorescence mo,oclonal antibody in TruCOUNT tube and detected by FACS CaUibur. Results were acquired by Multiset program and statistically analyzed by SPSS 12.0 software. 95% confidence interval was used to determine normal reference values. Results Women had a higher proportion of CD3 ^＋ cells and a higher proportion and absolute number of CD4 ^＋ cells. There were no significant differences in CD3 ^＋ cells＇ absolute number and in proportion and absolute number of CD8 ^＋ cells between genders. Increasing age was associated with a increased proportion and absolute number of CD4 ^＋ cells and a decreased proportion and absolute number of CD8^＋ cells, resulting in an age-related increase in the ratio of the CD4^＋ cells to CD8^＋ cells. Zhejiang health nomal values of T lymphocyte subsets were as follows： CD3^＋ （%）： 47.98 - 82.60; CD3^＋ （/μl）： 712.49-2367.55; CD8^＋ （%）： 14.24-41.48; CD8^＋ （/μl）： 219.14-1 109.81; CD4^＋ （%）： 24.15-51.66; CD4^＋ （/μl）： 383.61 - 1 345.60; CD4/CD8：0.70 - 3.25. Conclusion T lymphocyte subset levels varied slightly with age and gender. The reference range for nomal values of T lymphocyte subsets could be established in the same race living in the same area.
