高压氧对实验兔脑血管前后循环超微结构及ET-1、eNOS的影响

摘    要:目的 探讨高压氧(HBO)作用实验兔后颈内动脉和基底动脉血管壁超微结构的变化,以及对内皮素-1(ET-1)、内皮型一氧化氮合酶(eNOS)表达的影响.方法 20只兔按数字表法随机分为实验组和对照组,每组10只.实验组于0.22 MPa下暴露60 min,连续3 d;对照组正常饲养,未做任何处理.免疫组织化学染色法检测2组颈内动脉和基底动脉ET-1、eNOS的表达,电镜下观察2组颈内动脉和基底动脉超微结构的变化.结果 ET-1免疫阳性标记颗粒在颈内动脉表达较基底动脉明显增强,eNOS免疫阳性标记颗粒在基底动脉表达较颈内动脉增强.电镜下实验组颈内动脉血管壁内弹力膜疏松,脂质空泡形成,内皮细胞及细胞器呈轻度变性;基底动脉超微结构变化不明显.结论 实验兔在HBO作用下颈内动脉血管收缩因子ET-1表达增强,基底动脉血管舒张因子eNOS表达增强.超微结构的改变可能与血管收缩、血流量减少、脂质过氧化物堆积有关.
Abstract:
Objective To investigate changes in the ultrastructures and expression of endothelial nitric oxide synthase (eNOS) and endothelin-1 (ET-1) of internal carotid artery (ICA) and basal artery (BA) following hyperbaric oxygen (HBO).Methods Twenty rabbits were randomly divided into 2 groups: the experimental group and the control group. The rabbits in the experimental group were exposed to HBO at 2.2 ATA for 60 minutes for 3 days. The rabbits in the control group were normally fed without any treatment. The expressions of eNOS, ET-1 of ICA and BA in the 2 groups were compared by using immunohistochemical technique. Then, changes in the ICA and BA ultrastructures of rabbits in the two groups were examined under electron microscope.Results The expression of ET-1 in ICA was significantly higher than that in BA, while that of eNOS in BA was more intensive than that in ICA. When a comparison was made between the experimental group and the control group, the internal elastic membrane of ICA for the former was loosened, lipid vacuolization was present, and endothelial cells and organelles degenerated slightly. Nevertheless, no significant changes were noted in the ultrastructures of BA.Conclusions The expression of ET-1 in ICA increased and the expression of eNOS in BA intensified following HBO therapy. Changes in the ultrastructures might be correlated with vasoconstriction, reduction of blood flow and accumulation of lipid peroxides.
