黄连、生地及其配伍对2型糖尿病大鼠治疗作用的比较研究

摘    要:目的 观察比较黄连、生地及其配伍对2型糖尿病大鼠的治疗作用,探讨黄连丸方剂配伍的科学内涵.方法 采用高脂饲料喂养联合小剂量链脲佐菌素(STZ)尾静脉注射法诱导2型糖尿病动物模型,将模型动物随机分为黄连组、生地组、黄连+生地组、罗格列酮组及模型组,分别给予相应药物进行干预,另设正常对照组.干预4周后各组大鼠行口服葡萄糖耐量试验(oral glucose tolerance test,OGTT)和胰岛素释放试验(insulin release test,IRT),计算胰岛素曲线下面积(INSAUc)和胰岛素敏感指数(ISI);并检测血清三酰甘油(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)及高密度脂蛋白胆固醇(HDL-C).结果 ①与模型组比较,各治疗组各时间点血糖均显著降低,INSAUC显著增加,ISI显著升高(P<0.01);与黄连+生地组比较,黄连、生地各单药组各时间点血糖水平均显著高于复方组,INSAUC显著小于复方组,ISI显著低于复方组(P<0.01);②与模型组比较,各治疗组血清TG、TC、LDI-C显著降低,HDL-C显著升高,差异有统计学意义(P<0.01);与黄连+生地组比较,黄连、生地各单药组血清TG、TC、LDL-C显著高于复方组,HDI-C显著低于复方组(P<0.01).结论 黄连、生地及其配伍对2型糖尿病大鼠均有一定的治疗作用,但黄连生地配伍治疗效果优于各单味药,提示黄连生地配伍治疗2型糖尿病具有协同作用.

Comparative Study on Therapeutic Effect of Coptidis Rhizome,Rehmannia Dride Rhizome and Their Compatibility on Type 2 Diabetes Mellitus Rats

Abstract:Objective To observe the effects of coptidis rhizome,rehmannia dride rhizome and their compatibil ity on type 2 diabetes mellitus rats so as to explore the scientific connotation of the combinationof Rhizoma Coptidis Pill. Methods Type 2 diabetes mellitus animal model was induced by high fat fed in combination with low dose streptozotocin injected through vein tail. The diabetic rats were randomly divided into the following groups:copti dis rhizome group, rehmannia dride rhizome group, coptidis rhizome + rehmannia dride rhizome group, rosiglita- zone group and model group. Normal control group was also set up. After intervention for four weeks, oral glucose tolerance test(OGTT)and insulin release test(IRT)were done on each group,and the areas under insulin curve (INSAuc) and insulin sensitivity index(ISI)were calculated according to formulas. Serums TG, TC, LDI.-C, and HDI. C were detected. Results (D Compared to the model group, the levels of blood glucose in all treatment groups were significantly lower(P(0.01)and had significantly bigger INSAuc and higher ISI(P(0.01). Compared to coptidis rhizome + rehmannia dride rhizome group,coptidis rhizome group and rehmannia dride rhizome group had significantly higher level of blood glucose,smaller INSAuc and lower ISI(P〈0. 01). Compared to the model group,serum levels of TG,TC,LDL-C in all treatment groups were significant lower(P〈0.01), serum levels of HDL-C in all treatment groups were significant higher (P〈 0.01), Compared to coptidis rhizome + rehmannia dride rhizome group,eoptidis rhizome group and rehmannia dride rhizome group had significantly serum levels of TG,TC,LDL-C and lower serum levels of HDL-C. Conclusion Coptidis rhizome,rehmannia dride rhizome and their compatibility are effective on type 2 diabetes mellitus rats. The effect of their compatibility is better than the single-drug which infer that there was synergetic effect on treating type 2 diabetes mellitus rats for their compatibility.
