补肾活血方对激素性股骨头坏死大鼠BMSCs分化影响的研究

摘    要:目的 探讨补肾活血方含药血清对激素性股骨头坏死大鼠股骨近端骨髓间充质细胞体外分化的影响.方法 建立激素性股骨头坏死大鼠模型后,提取股骨近端骨髓单个核细胞,随机分为4组培养,空白血清对照组(A组)、补肾活血方含药血清低(B组)、中(C组)、高(D组)浓度组,分别加入重组人骨形态发生蛋白-2(rhBMP-2)诱导下的骨髓间充质细胞的培养体系,观察各组的细胞形态学、细胞内ALP含量及TGF-β基因表达.结果 补肾活血方含药血清能增强rhBMP-2的活性,促进股骨近端骨髓体外成骨细胞的分化增殖(P<0.01),促进分化中的骨髓间充质细胞表达TGF-β1 mRNA(P<0.01).结论 补肾活血方通过增强rhBMP-2活性及TGF-β1 mRNA的表达在骨坏死的修复中起关键作用.

Effect of Bushen Huoxue Decoction on the Osteogenic Differentiation of Bone Marrow Stromal Cells in the Steroid Induced Osteonecrosis of the Femoral Head in Rats

Abstract:Objective To detect the effects of Bushen Huoxue decoction on the osteogenic differentiation of bone marrow stromal cells in the steroid induced osteonecrosis of the femoral head in rats. Methods Established steroid induced necrosis of the femoral head rat model, extracted proximal femoral bone marrow mononuclear ceils. 40 male SD rats were randomly divided into four groups:Group A was cultured in blank serum,while,group B,C, and D were cultured in low,medium,and high concentration of Bushen Huoxue decoction serum respectively. The bone marrow stromal cells of all groups were cultured in osteoblasts induced by rhBMP-2. To observe the cell morphology,intracellular ALP content, and the expression of TGF-~I gene. Results Bushen huoxue decoction could enhance rhBMP-2 activity,improve differentiation and proliferation of the osteoblasts(P〈0.01),and facili- tate the expression of TGF-β1 mRNA of bone marrow stromal cells(P〈0.01). Conclusion Bushen Huoxue decoc- tion could enhance rhBMP-2 activity and enhance the expression of TGF-β1 mRNA, which plays a key role in the repair of bone necrosis.
