维拉帕米对缺血后大鼠视网膜电生理功能和节细胞代谢的影响

摘    要:本实验用闪光视网膜电图(FERG)、细胞色素氧化酶(CO)组织化学研究维拉帕米对缺血后大鼠视网膜的影响。结扎双侧颈总动脉,3h后松结复流。分别于缺血前15min和再灌流前1min腹腔注射维拉帕米(5mg/kg,25mg/m1/次),对照组腹腔注射等量生理盐水。动物分别存活1、3、7d,处死前复查FERGb波。视网膜铺片行CO组织化学反应,CO结果用计算机图像分析处理。结果显示:维拉帕米组与对照组缺血后第1dFERGb波恢复程度差异无显著性(P>005),而第3、7d,维拉帕米组FERGb波幅度恢复程度较对照组显著性增加(P<005),维拉帕米组高CO活性节细胞数密度大于对照组(P<005),高CO活性节细胞的灰度低于对照组(P<005)。说明维拉帕米对大鼠视网膜缺血后的节细胞具有保护作用,并促进视网膜功能的恢复。

Abstract:The effects of verapamil on the ischemic retina in an animal model was studied by means of cytochrome oxidase(CO) histochemistry and measuring the b wave of flash electroretinogram(FERG).Both left and right carotid arteries were clamped for 3h.Verapamil(5mg/ml in 2ml normal salime for verapamil-treated rats)or equal volume of the vehicle (for untreated animals)was injected intra-peritoneally 15 min before and 3h after the surgery.Then the carotid arteries were unclamped.After 1-,3-,7-day survival,the FERG-b wave was measured before the rats were sacrificed.The samples of retina were processed with CO histochemical reactions and the results were studied by image analysis system.The results showed that,comparing with the untreated animals,FERG b waves in the treated animals survived for 1 day were lower,but for 3 and 7 days were significantly higher than those in the controls .The high CO activity of retinal ganglionic cell density of the treated animals survived for 3 and 7 days was significantly higher,but the grey scale was significantly lower than the untreated animals.The present data suggest that verapamil may rescue post-ischemia retina ganglionlic cells,and also benifit to improve the retinal function.
