MicroRNA-218单核苷酸多态性与结直肠癌易感性的关系

摘    要:目的 探究microRNA-218单核苷酸多态性与结直肠癌易感性的关联。方法应用聚合酶链式反应-连接酶检测反应技术(PCR-LDR)对66例结直肠癌患者和124例健康对照者的microRNA-218单核苷酸的多态性进行分析,并采用二元逻辑回归分析评估此多态位点与结直肠癌易感性的相关性。同期对比检测microRNA-25、microRNA-143和 microRNA-106a的单核苷酸多态性。结果二元逻辑回归分析结果显示,相比AG和AA基因型,microRNA-218 rs11134527 的GG基因型显著增加结直肠癌的易感性(OR=2.31, P<0.05)。而microRNA-25 rs41274221、 microRNA-143 rs4705342和 microRNA-106a rs137881717的单核苷酸多态性与结直肠癌的易感性无关联(P>0.05)。结论MicroRNA-218 rs11134527单核苷酸多态性与结直肠癌易感性密切相关。

Abstract:Objective To investigate the function of microRNA-218 in colorectal cancer (CRC). Methods The polymerase chain reaction-ligation detection reaction (PCR-LDR) method was used to genotype the polymorphism of microRNA-218 in 66 CRC cases and 124 healthy controls. Furthermore, association between the risk for CRC and the four SNPs was evaluated via binary logistic regression analysis. Results Binary logistic regression analysis showed that GG genotype (OR=2.31, P<0.05) of microRNA-218 rs11134527 was significantly correlated to the increased risk of CRC, compared with the AG and AA genotype. However, no significant association was found between the SNP rs41274221 in microRNA-25 (P>0.05), the SNP rs4705342 in microRNA-143 (P>0.05) or the SNP rs137881717 in microRNA-106a (P>0.05) with CRC. Conclusion Micro218 rs11134527 is closely correlated to the susceptibility to CRC.
