Abstract:Objective: To detect the expression level of CXC chemokine ligand 14(CXCL-14) in non-small cell lung cancer(NSCLC) and analyze its clinical significance. Methods: A total of 260 patients with NSCLC who underwent surgical treatment at People′s Hospital of Zhengzhou from January 2017 to January 2019 were collected.The expression of CXCL-14 mRNA in NSCLC tissues,paracancerous tissues,NSCLC cells A549,h1975,PC-9 and BEAS-2B cells were detected;siCXCL-141(siCXCL-141 group),siCXCL-142(siCXCL-142 group) and si-RNA(control group) plasmids were transfected into A549 cells.The proliferation and apoptosis of transfected cells were detected. According to the average level of CXCL-14 expression,patients were divided into CXCL-14 high expression group and CXCL-14 low expression group,and the clinical characteristics of patients between different groups were analyzed. Results: The relative expression level of CXCL-14 mRNA in NSCLC tissues was(3.62±0.78),higher than that of adjacent tissues[(1.17±0.32)],the difference was statistically significant(t=22.440,P<0.001).The relative expression levels of CXCL-14 mRNA in A549,H1975 and PC-9 were(2.10±0.13),(1.84±0.21)and(1.47±0.08),respectively,which were higher than that in normal lung epithelial cells(0.69±0.08),and the difference was statistically significant(q=23.631,17.882,13.568,P<0.001).The relative expression levels of CXCL-14 mRNA and protein in siCXCL-141 group and siCXCL-142 group were lower than those in control group,and the difference was statistically significant (P<0.05). At 72 h,the OD values of cells in the siCXCL-141 group and siCXCL-142 group were(1.02±0.19) and(0.98±0.12),both of which were lower than those in control group(1.63±0.21),the difference was statistically significant(q=3.102,P=0.015;q=3.241,P=0.012);the apoptosis rates of siCXCL-141 group and si CXCL-142 group were (13.57±2.82)% and (14.41±2.14)%,which were higher than that of control group(5.58±0.69)%,the difference was statistically significant(q=4.774,P=0.009;q=7.031,P=0.002);there were more patients with poor differentiation,pathological stage Ⅱ,Ⅲ and lymph node metastasis in the CXCL-14 high expression group than those in the CXCL-14 low expression group. The number of 1-year progression-free survival patients was less than that in the CXCL-14 low expression group,the difference was statistically significant(P<0.05). Conclusion: The expression of CXCL-14 increases in NSCLC. Decreasing the expression of CXCL-14 can weaken cell proliferation and promote cell apoptosis. Increased expression of CXCL-14 can reflect the poor prognosis of patients,and is expected to become an effective target for the treatment of NSCLC.
