Effect of Dihydroartemisinin on the biological characteristics of HeLa cells

Abstract:Objective To investigate the effect and mechanism of dihydroartemisinin (DHA) on cell cycle, proliferation, apoptosis and invasion of HeLa cells. Methods HeLa cells were divided into three groups according to different concentrations of DHA, blank control group (PBS of equal volume), low concentration DHA group (50 μmol/L) and high concentration DHA group (400 μmol/L). Western blot was used to detect the expression of Caspase-3 and Bax protein, flow cytometry was used to detect cell cycle and apoptosis, plate cloning was used to detect cell proliferation, and transwell invasion was used to detect cell invasion. Results The expression of Bax, caspase-3 proteins in blank control group and low concentration DHA group were 0.48±0.07, 0.51±0.08 and 0.95±0.11, 1.03±0.12, respectively; and that in high concentration DHA group was 2.28±0.21, 2.35±0.23, respectively; which was significantly higher than those of the two groups (P<0.05). The cell cycle results showed that the cells in G0/G1 phase and S phase in blank control group, and low concentration DHA group were 53.52±3.98 and 32.84±3.18, and 61.64±3.92 and 22.47±2.31, respectively; and in high concentration DHA group were 76.48±5.26 and 13.25±1.22, respectively, which was blocked in G0/G1 phase (P<0.05), and the number of cells in S phase was significantly reduced (P<0.05). The apoptosis rate in high concentration DHA group was 20.41±0.92%, which was significantly higher than those in blank control group and low concentration DHA Group (2.84±0.61% and 10.36±0.63%, respectively, P<0.05). The number of cell clone formation (62.54±12.46) in high concentration DHA group was significantly lower than those in control group and low concentration DHA Group (188.56±14.84 and 119.56±14.62, respectively, P<0.05). The transwell cell count in high concentration DHA group (92.75±13.54) was significantly lower than those in control group and low concentration DHA Group (232.58±26.42 and 128.75±22.16, respectively, P<0.05). Conclusion DHA can increase the protein expression of Bax and caspase-3, inhibit the proliferation, promote cell apoptosis, reduce S-phase cells, and reduce cell invasion of HeLa cells. The effect is more significant at high concentration.
