建立鸭乙肝病毒感染肝郁脾虚病证动物模型的研究

摘    要:目的:建立鸭乙肝病毒感染肝郁脾虚病证动物模型。方法:50只1日龄雏鸭随机分为4组:病毒组、模型组雏鸭通过腹腔注射DHBV-DNA强阳性血清使其感染病毒。后模型组、限制组用柔软细绳条束缚雏鸭双翅及双下肢、置于空地强行放牧、隔日喂食等方法建立肝郁脾虚模型。空白组及病毒组攻毒后正常饲养。观察4组肝脏病理学检查结果、啄癖率、进食时间、强行下水时间、血液流变学指标、肝功能指标、免疫学指标变化。结果:除空白组外,其余各组肝细胞均见有轻度变性,模型组、病毒组更甚。模型组、限制组发生啄癖的雏鸭数、进食时间、强行下水时间均明显较空白组、病毒组多或短,与空白组比较,差异有非常显著性意义(P〈0.01)。病毒组、模型组血液流变学各项指标均明显较空白组、限制组为高,与空白组比较,差异均有显著性意义(P〈0.05)。肝功能指标病毒组、模型组、限制组均明显升高,与空白组比较,差异均有显著性意义(P〈0.05)。限制组、病毒组与模型组比较,差异均有显著性意义(P〈0.05)。免疫学指标病毒组、模型组、限制组均较高,与空白组比较,差异均有显著性意义(P〈0.05)。结论:慢性乙型肝炎肝郁脾虚型的临床表现可以部分通过雏鸭模型模拟,成为中西医结合的病证动物模型。

Abstract:Objectlve:To establish the duckling models of liver-stagnation and spleen-deficiency infected by Duck Hepatitis B Virus(DHBV). Methods: Fifty one-day old ducklings were randomized into 4 groups: virus group and model group received DHBV-DNA strong positive serum by intraperitoneal injection to be infected. Then model group and restrict group established models of liver-stagnation and spleendeficiency by soft string bingding up both wings and lower limbs of ducklings, arbitrary herding on open space, feeding every other days. While blank group and virus group were normal fed. The changes of hepatic histopathologic finding, rate of feather pecking, intaking time, arbitrary swimming time, hemorheology index, liver-function index, immunology index were observed. Result: The hepatocyte cells were degenerated except those of blank group, model group and virus group being much more remarkable. The amount of feather pecking ducklings, intaking time, arbitrary swimming time in model group and restrict group were more large or short than those in blank group and virus group, the difference being very significant as compared with those in blank group(P〈0.01). The hemorheology indexs in virus group and model group were higher than those in blank group and restrict group, the difference being significant as compared with those in blank group(P〈0.05). The liver-function indexs were increased except in blank group(P〈0.05, as compared with those in blank group). The difference being significant as restrict group and virus group compared with model group (P〈0.05). The immunology indexs were increased except in blank group(P〈0.05, compared with those in blank group).Conclusion: The manifestations of duckling models of hver-stagnation and spleen-deficiency infected by DHBV can be partially simulated to establish the desease and syndrome animal model integrated traditional Chinese and western medicine.
